鲑鱼降钙基因向烟草遗传转化的初步研究

摘要:
报道了鲑鱼降钙素基因植物表达载体的构建及其向烟草的遗传转化。由鲑鱼降钙素的链霉菌表达载体PMS680,通过限制性内切酶BamH I和HindⅢ双酶切后将其克隆至中间载体PART7,获得中间表达载体PART7-SCT,从而给目的基因加上启动和终止子,将限制性内切酶Notl酶切PART7-SCT得到的带有启动子和终止子的目的基因片段克隆至载体PAPT27,得到鲑鱼降钙素基因的植物表达载体PART27-SCT。通过农杆菌介导的叶盘法将其转至烟草,经PCR检测巳得到转基因植株。

A study on the transformation of salmon calcitonin gene into tabacco

Abstract:
This paper reported the construction of the recombinant expression plasmids of salmon calcitonin gene for the first time.A gene coding for salmon calcitonin (sCT) was obtained from its streptomyces vector pMS680 by digestion of restriction enzyme BamHⅠand Hin dⅢ.The gene coding was cloned into vector pART27 to generate plant expression vector pART27-sCT.The infection of Agrobacterium tumefaciens was used to transform tobacco plant,PCR of transformed tobacco DNA demonstrated the integrarion of sCT expression vector into the transgenic tobacco genome.
