农标菌介导的花椰菜遗传转化体系研究

摘要:
以花椰菜栽培品种“春秋”无菌苗的子叶和下胚轴为外植体,在携带CaMV Bari-1基因Ⅵ的根癌农杆菌菌种GV3101的介导下,对影响花椰菜遗传转化的诸因素进行了研究,建立了较为完善的转化体系,其转化率分别达2.31%和2.56%。在这一转化体系中,首先将过夜培养的农杆菌用MS无机盐液体培养基稀释5倍或10倍,感染经2~3 d预培养的子叶和下胚轴外植体30s,然后进行黑暗共培养1 d,外植体经表达灭菌后转接到含500 mg/L羧苄青霉素的分化培养基上。3周后,将分化的不定芽切成0.5 cm见方的小块,接种在选择培养基上,经筛选的转化芽再诱导成苗。

Studies on genetic transformation system of cauliflower mediated by Agrobacterium tumefaciens
Abstract:
Effects of some factors on transformation were studied with cotyledon and hypocotyl from Brassica Oleracea Var.botrytis cv.“ChunQiu” by Agrobacterium tumefaciens strain GV3101 containing CaMV Bari-1 gene Ⅵ.Genetic transformation system was established and the transformation frequency was 2.31% and 2.56% respectively.In this transformation system,agrobacterium grown overnight was diluted at a ratio of 1∶5 or 1∶10 with MS salts liquid medium,and cotyledons and hypocotyls precultrued for 2or 3 days were inoculated for 30 s in it,and then cocultivated for 1 d in the dark.Inoculated explants were placed on the differentiation medium containing Cb 500 mg/L after sterilization.After 3 weeks cut the bud into pieces of 0.5 cm3 and place them on the selective medium,and the transformation bud was regenerated into plantlet.
