CP基因转化的线辣椒抗卡那霉素和抗CMV特性的遗传

摘要:
利用根癌土壤标菌,采用叶盘法转化线辣椒(Capsicum annuum var.longunt)优良品种陕8212,建成了能同时表达CMV和TMV外壳蛋白基因(CP基因)的T1代纯合系。以其自交T2~T4代,以及T2代植株与未转化陕8212杂交的F1代和F2代群体为试材,研究了抗卡那霉素和抗黄瓜花叶病毒(CMV)特性的遗传递规律。结果发现,抗卡娜霉素标记基因和抗病性基因在自交和杂交各代都能稳定地高效表达;两者在自交各代纯合;在杂交F1代抗药性和抗病性都表现为显性;在杂交F2代,抗药植株对敏感植株,以及抗病植株对感病植株的分离比都符合3∶1的理论比例。表明抗药性和抗病性均为显性单基因遗传。

Inheritance of kanamycin-resistance and CMV-resistance in transgenic chili pepper expressing CMV-CP and TMV-CP genes
Abstract:
Both CMV-CP and TMV-CP genes were transferred into cultivar“Shaan 8212” of chili peper (Capicum annuum var.longunt)mediated by Agrobacterium tumefacience.Selfing generation T2,T3 and T4 from homozygous T1 lines and F1,F2 of cross T2/Shaan 8212 were tested and analysed to reveal inheritance of resistance to Kanamycin or to CMV.The results showed that resistance steadily and strongly expressed int tested populations.T2,T3 and T4 were in homozygous condition showing high resistance ot Kanamycin and to CMV.The resistance was dominant over sensitivity to Kanamycin or susceptibility to CMV in F1 progenies.F2 segregated to fit 3∶1 ratio.It was concluded that resistance to Kanamycin and resistance to CMV were conditioned by single dominant genes.
