| LALBA基因多态性对绵羊泌乳性状的影响 |

| 【 目的】筛选 α-乳清蛋白(alpha-lactalbumin,LALBA)基因的单核苷酸多态性(single nucleotide poly‐ morphisms,SNP)位点,并探究该基因多态性对绵羊泌乳性状的影响,为绵羊分子遗传育种研究提供理论参考。【 方 法】以437只奶绵羊为试验对象,采用Sanger测序筛选LALBA基因SNP位点,并用五引物扩增受阻突变体系(pentaprimer amplification refractory mutation system, PARMS)对SNP进行分型,最后用一般线性混合效应模型分析基因 型、等位基因和单倍型对绵羊泌乳性状的影响。【 结果】 在绵羊LALBA基因5'非翻译区(5' untranslated region,5' UTR)、第1外显子与第1内含子区域各筛选到1个SNP位点,分别为 c.-89 G/A、c.106 C/T和c.160-13 G/A。相关性分析发现,在c.-89 G/A位点,GG型个体的乳脂率显著高于AA型(P<0.05),AA型个体的乳糖率显著高于GA和GG型个体;等位基因G与较高的乳脂率相关,与较低的乳糖率和非脂乳固体含量相关。在c.106 C/T 位点,CC型个体的乳脂率显著高于TT型,TT型的乳糖率显著高于CT和CC型;等位基因C与较低的乳蛋白率、乳糖率和 非脂乳固体含量显著相关(P<0.05)。在c.160-13 G/A位点,GG型个体的乳电导率和pH值显著高于GA型;等位基因A与较低的乳电导率和pH值相关。在发现的3种单倍型中,CCGG单倍型组合的乳脂率显著高于TTAA型(P<0.05)。TTAA单倍型组合的乳糖率显著高于CCGG和CTGA型。【 结论】在绵羊LALBA基因中发现了3个SNP位点,其中c.-89 G/A和c.106 C/T影响了乳脂率和乳糖率,c.160-13 G/A与电导率和pH值相关。 |

| 【Objective】This study aims to screen single nucleotide polymorphisms(SNP) in the alpha-lact‐albumin(LALBA) gene and explore the effects of its polymorphism on the lactation traits of sheep,providing a theoretical foundation for molecular genetic research on this species.【Method】A total of 437 dairy sheep were used as experimental subjects. SNP in the LALBA gene were screened by Sanger sequencing and genotyped us‐ing the penta-primer amplification refractory mutation system(PARMS). Subsequently,a general linear mixed effects model was applied to analyze the effects of genotypes,alleles,and haplotypes on the lactation traits of sheep.【Result】Results showed three SNP in the LALBA gene:c.-89 G/A in the 5' untranslated region(5' UTR),c.106 C/T in the exon 1,and c.160-13 G/A in the intron 1. Correlation analysis revealed that for c.-89 G/A locus,individuals with the GG genotype exhibited a significantly higher milk fat percentage than those with the AA genotype(P<0. 05),whereas individuals with the AA genotype showed a significantly higher lactose percentage than those with the GA and GG genotypes. Allele G was associated with a higher milk fat percentage,but a lower lactose percentage and non-fat solid content. For c. 106 C/T locus,individuals with the CC genotype showed a significantly higher milk fat percentage than those with the TT genotype,whereas in‐dividuals with the TT genotype displayed a significantly higher lactose percentage than those with the CT and CC genotypes. Allele C was related to lower milk protein percentage,lactose percentage,and non-fat solid content(P<0. 05). For c.160?13 G/A locus,individuals with the GG genotype demonstrated significantly higher electrical conductivity and pH values than those with the GA genotype. Allele A was linked to lower electrical conductivity and pH values. Among the three haplotypes identified,the CCGG haplotype exhibited a signifi‐cantly higher milk fat percentage than the TTAA haplotype(P<0. 05),whereas the TTAA haplotype showed a significantly higher lactose percentage than the CCGG and CTGA haplotypes.【Conclusion】Three SNP loci were identified in the LALBA gene of the sheep. Among them,c.?89 G/A and c. 106 C/T affected the milk fat content and lactose content;while c. 160?13 G/A was related to electrical conductivity and pH value. |
