增补UV‑A对银杏生长生理及黄酮积累的影响

摘要:
【 目的】探究夜间增补不同强度UV-A对银杏生长生理以及类黄酮代谢的影响,选择适宜的夜间补光光质及技术提高银杏药用经济价值。【 方法】以1年生半同胞金叶银杏与绿叶银杏盆栽实生苗为研究对象,分别设置对照组(白光,300 μmol/(m2·s))和试验组,其中试验组在白天正常照射结束后,夜间增补强度分别为15,30和45 μmol/(m2·s)390 nm的UV-A紫外光,测定其不同生长期生物量、矿质元素和初生代谢物含量、抗氧化酶和苯丙烷生物合成酶活性以及总黄酮含量,分析夜间增补不同光照强度UV-A对银杏生长及生理代谢的影响。【 结果】①在夜间增补30 μmol/(m2·s)UV-A处理下,与对照组相比,绿叶银杏地上部生物量在快速生长期显著增加55.57%,在封顶期 根冠比最小,为对照的64%,生长状况最佳;生物量与矿质元素含量呈显著正相关。②在夜间增补30 μmol/(m2·s)UV-A处理下,封顶期金叶和绿叶银杏叶片的可溶性蛋白含量显著升高,分别为对照组的2.02和1.90倍。在封顶期,金叶银杏和绿叶银杏分别在夜间增补30和45 μmol/(m2·s)处理下POD活性最高,相较于对照分别增长19.8%和18.4%,且均显著大于对照组。③苗木封顶期,绿叶银杏的苯丙烷生物合成酶活性和叶片总黄酮含量在夜间增补30 μmol/(m2·s)的UV-A处理下最高,其总黄酮含量是对照的2.07倍。④模糊隶属函数法综合评价结果显示,在夜间增 补30和45 μmol/(m2·s)处理下银杏植株的隶属度均值均较高,表明该处理下叶片黄酮药用价值最佳。【 结论】在白天正常照射条件下,夜间增补30和45 μmol/(m2·s)的UV-A最有利于银杏幼苗生长和叶片黄酮积累,提升药用价值。

Effects of nighttime supplemental UV⁃A at different intensities on the growth physiology and flavonoid accumulation of Ginkgo biloba

Abstract:
【 Objective】The study aimed to investigate the effects of different intensities of nighttime supple?mental UV-A on the growth,physiology,and flavonoid metabolism of Ginkgo biloba,and select optimal nighttime supplemental light quality and techniques to enhance its medicinal and economic value.【Method】One-year-old half-sibling potted seedlings of golden-leaf G.biloba and green-leaf G.biloba were used as materials.The seedlings were divided into control and experimental groups. After normal daytime light exposure,the experimental groups were exposed to supplemental nighttime 390 nm UV-A radiation at three different intensities:15,30,and 45 μmol/(m2·s). During different growth stages,measurements were taken of biomass,mineral elements,primary metabolites,antioxidant-related enzymes,phenylpropanoid biosynthesis-related enzymes,and total flavonoids. The effects of nighttime supplemental UV-A at different intensities on the growth and metabo-lism of G.biloba were analyzed.【 Result】①When supplemented with nighttime UV-A irradiation at an inten-sity of 30 μmol/(m2·s),the aboveground biomass of green-leaf G.biloba significantly increased by 55.57% during the rapid growth period compared with the control group. At the terminal bud set stage,the root-to-shoot ratio was at its lowest,64% of that in the control group,indicating the optimal growth conditions. The biomass showed significant postive correlation with the mineral elements content. ②Under supplemental nighttime UV-A treatments of 30 μmol/(m2·s),the soluble protein content in the leaves of golden- and green-leaf G.biloba at the terminal bud set stage significantly increased,reaching 2.02-fold and 1.90-fold that of the control,respec-tively. At this stage,golden-leaf G.biloba exhibited the highest POD activity under the 30 μmol/(m2·s) treat?ment,while green-leaf G.biloba showed the highest activity under the 45 μmol/(m2·s) treatment,with increases of 19.8% and 18.4% compared with the control,respectively. Both values were significantly higher than those of the control. ③During the terminal bud set stage,the phenylpropanoid biosynthesis enzyme activity and total flavonoid content in the leaves of green-leaf G. biloba reached their highest levels under supplemental night-time UV-A radiation of 30 μmol/(m2·s),with the total flavonoid content being 2.07 times that of the control. ④ A comprehensive evaluation using the fuzzy membership function method revealed that the mean membership de-grees of G.biloba plants were relatively high under supplemental nighttime UV-A treatments of 30 and 45 μmol/(m2·s),indicating that these treatments yielded the optimal medicinal value of leaf flavonoids.【Conclusion】Under normal daytime irradiation conditions,supplemental nighttime UV-A radiation at 30,45 μmol/(m2·s) is most conducive to the growth of G.biloba seedlings and the accumulation of flavonoids in their leaves,thereby enhancing their medicinal value.
