皮下埋植褪黑素对西门塔尔肉牛繁殖性能、血清激素、抗氧化和免疫指标的影响

摘要:
【 目的】探究褪黑素(MT)埋植对西门塔尔肉牛繁殖性能、血清激素、抗氧化和免疫指标的影响。【 方法】选择64头繁殖机能正常的母牛,随机分为4组,每组16头,采用CIDR+GnRH+PG+PG+GnRH方案进行同期排卵-定时授精处理,在各组母牛第1次注射GnRH的同时,分别颈部皮下埋植 0(对照组)、60(1组)、120(2组)和180(3组) mg/头MT,在第2次注射GnRH后8 h检测血清中MT、促卵泡素(FSH)、促黄体素(LH)、雌二醇(E2)和抗氧化指标的水平,人工授精后14和30 d检测血清MT、干扰素-tau(IFNT)、孕酮(P4)、抗氧化和免疫指标的水平,人工授精后45 d进行妊娠诊断,统计受胎率,母牛分娩后统计产犊率。【 结果】①与对照组相比,皮下埋植 MT 未显著提高受胎率和产犊率(P>0.05)。②与对照组相比,试验组母牛排卵前和妊娠早期血清生殖激素水平呈增加趋势,试验2组效果最好,其排卵前母牛血清MT、FSH、LH和E2水平显著升高(P<0.05),妊娠14和30 d试验2组母牛血清MT、P4和IFNT水平显著升高(P<0.05)。③与对照组相比,试验组母牛排卵前和妊娠早期血清抗氧化指标呈增加趋势,试验2组效果最好,其母牛血清总抗氧化能力(T-AOC)及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)活性显著升高(P<0.05),而血清丙二醛(MDA)含量显著降低(P<0.05)。④ 与对照组相比,试验组母牛妊娠早期血清免疫指标呈增加趋势,试验2组效果最好,其母牛血清免疫球蛋白 G( IgG)、IgA和IgM水平显著升高(P<0.05)。【 结论】皮下埋植120 mg/头MT能够提高西门塔尔肉牛母牛血清 FSH、LH、E2、P4和IFNT含量,增强血清抗氧化和免疫功能,使受胎率升高25%。

Effects of melatonin implantation on reproductive performance,serum hormone levels,antioxidative and immune indexes of Simmental cows

Abstract:
【 Objective】This research aims to explore the effects of melatonin(MT) implantation on the reproductive performance,serum hormones,and antioxidant indexes of Simmental cows.【Method】64 Simmental cows with normal reproductive function were randomly divided into 4 groups, with 16 cows in each group. The CIDR+GnRH+PG+PG+GnRH protocol was adopted for synchronized ovulation and fixed-time insemination. At the time of the first GnRH injection,cows in each group were subcutaneously implanted in the neck with 0 mg(the control group),60 mg(Group 1),120 mg(Group 2),and 180 mg(Group 3) of MT. The serum levels of MT,follicle stimulating hormone(FSH),luteinizing hormone(LH),estradiol(E2),and antioxidant indexes were detected 8 hours after the second GnRH injection. 14 and 30 days after artificial insemination,the serum levels of interferon tau(IFNT),progesterone(P4),and antioxidant indexes were measured. Pregnancy diagnose was carried out 45 days after artificial insemination and the conception rate of cows was calculated,and calving rate was calculated after calving.【Result】The results showed that:① Compared with the control group,subcutaneous implantation of melatonin(MT) did not significantly increase the conception rate and calving rate (P>0.05),but there was an increasing trend in the conception rate,with the highest conception rate observed in group 2. ②Compared with the control group,there was an increasing trend in the serum reproductive hormone levels of trial group cows both before ovulation and during the early stage of pregnancy. Group 2 exhibited the most favorable results,with significant increases in serum melatonin (MT),follicle-stimulating hormone (FSH),luteinizing hormone(LH),and estradiol(E2) levels before ovulation(P<0.05). 14 and 30 days after artificial insemination,the levels of serum MT,progesterone(P4),and interferon-tau( IFNT) in cows from Group 2 were also significantly elevated(P<0.05). ③ Compared with the control group,there was an increasing trend in the serum antioxidant indexes of trial group cows both before ovulation and during the early stage of pregnancy. The best results were observed in Group 2,with significant elevations in the serum total antioxidant capacity(T-AOC) and the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and catalase(CAT) in cows(P<0. 05),and significant decrease in the serum malondialdehyde( MDA) content (P< 0.05). ④ Compared with the control group,there was an increasing trend in the serum immune indexes of the trial group cows during the early stage of pregnancy. The best results were observed in Group 2,with significant elevations in the levels of serum immunoglobulin G( IgG),IgA,and IgM(P<0.05).【Conclusion】In summary, subcutaneous implantation of 120 mg MT can improve serum FSH,LH,E2,P4 and IFNT levels of Simmental beef cow,enhance serum ntioxidant and immune function of Simmental beef cow,and increase conception rate by 25%.
