基于小RNA测序的香瓜茄病毒病病原鉴定

摘要:
【目的】利用小RNA(sRNA)测序技术和RT-PCR技术检测我国香瓜茄主要种植区的香瓜茄病毒病病原种类及其侵染情况,为香瓜茄病毒病害的精准防控提供依据。【方法】以田间调查采集的香瓜茄病样为材料,通过小RNA测序技术和RT-PCR技术,挖掘我国6个香瓜茄种植区的主要病毒种类,并分析香瓜茄病毒在不同地区的分布及其复合侵染情况。【结果】sRNA测序结果表明,6个香瓜茄种植区比对得到的病毒属于8个病毒科,主要以(+)ssRNA为主,其中勐腊、西宁、民和、民勤 4个地区香瓜茄病毒病的主要病原为番茄花叶病毒(tomato mosaic virus,ToMV),石林地区香瓜茄病毒病的主要病原为辣椒叶脉斑驳病毒(chilli veinal mottle virus,ChiVMV),玉门地区香瓜茄病毒病的主要病原为番茄斑萎病毒(tomato spotted wilt virus,TSWV)。RT-PCR验证结果表明,香瓜茄普遍受番茄花叶病毒、马铃薯S病毒(potato virus S,PVS)和烟草花叶病毒(tobacco mosaic virus,TMV)侵染,共存在9种复合侵染类型,且以双重侵染为主,占比高达42.36%。【结论】侵染香瓜茄的病毒多属于(+)ssRNA病毒,首次在香瓜茄中发现TVBMV、ChiVMV和TSWV的侵染。云南省香瓜茄病毒病害最为严重,甘肃省香瓜茄病毒病害较轻,国内香瓜茄主要受到ToMV、 PVS 和TMV的侵染,且以复合侵染为主。

Pathogen identification of Solanum muricatum viral diseases by small RNA sequencing

Abstract:
【Objective】This study aimed to identify the types and infection status of viruses in the main pepino (Solanum muricatum) cultivation areas in China using small RNA (sRNA) sequencing and RT-PCR techniques,to provide a basis for the precise prevention and control of viral diseases in pepino.【Method】With pepino disease samples collected from field surveys as materials,small RNA (sRNA) sequencing technology and RT-PCR technology were applied to identify the main virus types in six pepino planting regions in China.The distribution of pepino viruses in different regions and the occurrence of mixed infections were analyzed.【Result】sRNA sequencing results showed that viruses identified in the six regions belonged to eight virus families,with (+)ssRNA viruses being predominant.In Mengla,Xining,Minhe and Minqin,the primary virus infecting pepino was tomato mosaic virus (ToMV).In Shilin,the main virus was chilli veinal mottle virus (ChiVMV),and in Yumen,it was tomato spotted wilt virus (TSWV).RT-PCR verification confirmed the common infection of pepino by three viruses:tomato mosaic virus (ToMV),potato virus S (PVS),and tobacco mosaic virus (TMV).Nine types of mixed infections were identified,with dual infections being the most prevalent,accounting for 42.36% of all infections.【Conclusion】Most viruses infecting pepino were (+)ssRNA viruses.This study provided the first report of the infection of pepino by TVBMV,ChiVMV,and TSWV.Pepino viral diseases were most severe in Yunnan Province,while Gansu Province experienced lighter disease outbreaks.The main viruses affecting pepino in China were ToMV,PVS and TMV,with mixed infections being the primary contributor to the diseases.
