外源褪黑素对葡萄幼苗铜毒害的缓解效应研究

摘要:
【目的】探究不同浓度外源褪黑素(MT)对葡萄幼苗铜毒害的缓解效应及生理机制,为葡萄栽培和生产提供理论依据。【方法】以山西自育葡萄品种‘秋红宝’为试材,以100 μmol/L CuSO4模拟铜毒害,通过叶面喷施50,100,200 μmol/L MT,分析外源MT对葡萄种子萌发及幼苗生长的影响。【结果】喷施MT可显著提高铜毒害下种子发芽率和发芽势,葡萄幼苗的生长量和叶绿素含量显著增加,抗氧化酶活性和渗透调节物质含量显著提高,葡萄幼苗叶片和根系的丙二醛(MDA)含量显著降低,相对电导率也随之降低,葡萄叶片的上下表皮厚度增加。【结论】外源MT可以有效缓解‘秋红宝’葡萄铜毒害,且褪黑素浓度在100 μmol/L时缓解效果最佳。

Studies on mitigating effect of exogenous melatonin on copper toxicity in grape seedlings

Abstract:
【Objective】The present study aims to elucidate the ameliorative impacts and physiological mechanisms of exogenous melatonin (MT) on the adverse effects of copper toxicity in grape seedlings, in order to provide theoretical insights for enhancing grape cultivation and production practices.【Method】The autochthonous grape variety ‘Qiuhongbao’ from Shanxi Province was selected as the experimental materials.Copper toxicity was simulated using 100 μmol/L CuSO4.50,100,and 200 μmol/L exogenous MT was applied to the leaves of the grape seedlings, and the effects of MT on seed germination and seedling growth were analyzed. 【Result】The application of MT enhanced the germination rate and vigor of seeds under copper toxicity conditions.Moreover,MT enhanced the growth and chlorophyll content of grape seedlings.The activities of antioxidant enzymes were enhanced,along with an increase in the levels of osmotic adjustment substances,indicating improved stress tolerance.Additionally,the levels of malondialdehyde (MDA) in leaves and roots of grape seedling were reduced and the relative electrical conductivity was decreased,suggesting improved cellular membrane stability.Furthermore,the thickness of the upper and lower epidermis in grape leaf was increased,with enhanced structural integrity.【Conclusion】Exogenous MT effectively mitigates the adverse effects of copper toxicity in ‘Qiuhongbao’ grape seedlings,with the optimal ameliorative effect at 100 μmol/L.
