环割对烤烟上部叶化学成分、感官质量及代谢物的影响

摘要:
【目的】探究环割对烤烟上部烟叶品质的影响,为提高上部烟叶可用性栽培措施改良提供参考。【方法】选取云烟100作为试验品种,设置不环割、环割2个田间处理,分析环割对烤烟上部叶化学成分含量、感官质量及代谢物的影响,并结合主成分分析(PCA)及正交偏最小二乘 判别分析(OPLS-DA)法对代谢物进行分析,以变量投影重要度(VIP)≥1.0,差异倍数FC值≥1.5或FC值≤0.667且P≤0.01筛选差异代谢物,研究环割对烤烟上部叶代谢物的影响。【结果】①环割处理后上部烟叶还原糖、总糖和有机酸含量得到一定提升,含氮物质含量明显减少,钾及多酚含量明显增加,化学成分协调性更佳,感官质量得到较大提升。②不同处理烤烟上部烟叶中共检测到405个挥发性代谢物,可分为14类;存在显著性差异的代谢物共63个,以萜类、酯类和酮类代谢物为主,共注释到6条代谢通路,显著富集于倍半萜和三萜生物合成通路。③环割处理对烤烟上部烟叶中萜类化合物代谢影响较为显著,松香芹酮、β-倍半萜类烯、愈创木酚、大马士酮等具有香味的代谢物与感官质量指标得分呈高度正相关,且其含量在环割处理的上部烟叶中较高。【结论】环割可提高烤烟上部叶化学成分协调性,有利于提高萜类物质代谢,进而改善感官质量,可作为提高烤烟上部烟叶可用性的改良措施。

Effects of girdling on the chemical composition,sensory quality and metabolites of upper flue-cured tobacco leaves

Abstract:
【Objective】This research aimed to explore the effect of girdling on the quality of upper leaves of flue-cured tobacco,to provide reference for improving cultivation measures to enhance the usability of upper tobacco leaves.【Method】Yunyan 100 variety was selected for the experiment.Two field treatments were set:no girdling and girdling.The effects of girdling on the chemical composition,sensory quality,and metabolites of the upper tobacco leaves were analyzed.Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were used to select differential metabolites with variable importance in projection (VIP)≥1.0,fold change (FC)≥1.5 or FC≤0.667,and P≤0.01,to study the impact of girdling on the metabolites of upper tobacco leaves.【Result】①After girdling,the contents of reducing sugars,total sugars,and organic acids in the upper leaves were increased,and the contents potassium and polyphenol were significantly increased,while nitrogen content was significantly decreased,indicating better chemical composition coordination and improved sensory quality.②A total of 405 volatile metabolites were detected in the upper tobacco leaves in different treatments,and they were categorized into 14 classes.Significant differences were observed in 63 metabolites,primarily terpenes,esters,and ketones,which annotated 6 metabolic pathways,which were mainly the pathway of sesquiterpenes and triterpenes biosynthetics.③Girdling significantly affected the metabolism of terpene compounds in the upper tobacco leaves,with compounds such as pine carvone,β-sesquiterpenoids, guaiacol,and damascenone showing a highly positive correlation with sensory quality scores,and higher contents in the girdling treatment.【Conclusion】Girdling can enhance the chemical composition coordination of upper flue-cured tobacco leaves,promote terpene metabolism, and improve sensory quality,which can be used as a potential method to enhance the usability of upper tobacco leaves.
