新疆大豆根腐线虫病病原鉴定及致病性测定

摘要:
【目的】明确从新疆喀什地区泽普县大豆根系和根际土壤样本中采集到的一种根腐线虫种类及其对大豆的致病性,为识别和防治大豆根腐线虫病提供指导。【方法】使用贝曼漏斗法从采集的大豆根系和根际土壤中分离线虫,并在胡萝卜愈伤组织上进行纯化培养,通过形态学和分子生物学相结合的方法鉴定分离到的根腐线虫种类,采用盆栽接种试验评估该根腐线虫种群对大豆的致病性及其对大豆生长的影响。【结果】形态特征分析结果显示,采集到的大豆根腐线虫种群与咖啡短体线虫(Pratylenchus coffeae)的形态特征较为一致。分子鉴定结果显示,该大豆根腐线虫种群的rDNA 28S D2-D3区序列和rDNA-ITS序列与NCBI数据库中咖啡短体线虫的序列相似度最高,分别为98.59%~99.87%和97.83%~99.10%。系统发育分析结果表明,该大豆根腐线虫种群与其他已鉴定的咖啡短体线虫种群处于同一高度支持的分支上。盆栽接种试验结果显示,接种该根腐线虫种群60 d后,大豆植株地上部矮小,长势较弱,茎秆较细,根系减少,且根系出现褐色坏死病斑,甚至腐烂症状,该线虫在大豆根际的繁殖系数达到4.03。【结论】从新疆喀什地区泽普县采集的大豆根腐线虫种类为咖啡短体线虫,其对大豆具有较强的致病性。

Identification and pathogenicity determination of a root-lesion nematode on soybean in Xinjiang

Abstract:
【Objective】This research aimed to identify a root-lesion nematode that was collected from the root and rhizosphere soil samples of soybean in Zepu County,Kashgar Prefecture,Xinjiang,and to determine its pathogenicity to soybean,in order to provide a guidance for the identification and prevention of root-lesion nematode disease on soybean.【Method】Nematodes were isolated from the collected soybean roots and soybean rhizosphere soil with the Baermann funnel method,and then purified and cultured on the carrot callus tissue.The isolated root-lesion nematode species was identified by the combination of morphological and molecular biological methods.Pot inoculation test was used to evaluate the pathogenicity of the root lesion nematode population to soybean and its effect on soybean growth.【Result】 The results of morphological analysis showed that the soybean root lesion nematode population collected in this study was similar to Pratylenchus coffeae.Molecular identification results indicated that the rDNA 28S D2-D3 region sequence and rDNA-ITS sequence of the soybean root-lesion nematode population showed the highest similarity with the sequences of P.coffeae in the NCBI database,which were 98.59%-99.87% and 97.83%-99.10%,respectively.Phylogenetic analysis showed that the soybean root-lesion nematode population was in the same highly supported clade with other identified populations of P.coffeae.The results of pot inoculation test showed that 60 days after inoculation with the root-lesion nematode population,soybean plants exhibited features of stunted growth,weak vigor,thin stems,reduced root systems,brown necrotic lesions on roots,and even symptoms of rot.The reproductive factor of the nematode in the soybean rhizosphere reached 4.03.【Conclusion】The species of root-lesion nematode collected from the rhizosphere of soybean in Zepu County,Kashgar Prefecture,Xinjiang is P.coffeae,which shows high pathogenicity to soybean.
