E3泛素连接酶TaRING2基因在小麦抗条锈菌过程中的功能分析

摘要:
【目的】研究E3泛素连接酶TaRING2基因在小麦抗条锈病过程中发挥的作用,为小麦条锈病的精准防控提供分子理论依据。【方法】以小麦品种水源11和条锈菌生理小种CYR23、CYR31为试验材料,通过实时荧光定量PCR、烟草亚细胞定位、病毒介导的基因沉默技术(BSMV-VIGS)等研究TaRING2基因在小麦与条锈菌互作中的作用。【结果】TaRING2基因全长1 473 bp,编码490个氨基酸。TaRING2蛋白相对分子质量为 51.8 ku,等电点为 9.15。亚细胞定位分析结果表明,TaRING2定位于烟草细胞膜和细胞核中。qRT0PCR分析结果显示,在接种后12和24 h,与小麦和条锈菌亲和体系相比,TaRING2在小麦和条锈菌互作的非亲和体系中极显著上调表达,相对表达量约为小麦与条锈菌亲和体系的10倍。利用病毒介导的基因沉默技术(BSMV-VIGS)瞬时沉默TaRING2基因,结果显示沉默了TaRING2基因的小麦植株对条锈菌的抗病性减弱,条锈菌的产孢量明显增加。组织学观察发现,在沉默了TaRING2基因的小麦植株中,条锈菌菌丝发育速度加快。【结论】E3泛素连接酶TaRING2基因正调控小麦抗条锈菌进程。

Functional analysis of E3 ubiquitin ligase TaRING2 gene in wheat resistance to stripe rust fungus

Abstract:
【Objective】This research aimed to investigate the role of wheat E3 ubiquitin ligase TaRING2 in wheat resistance to stripe rust,and to provide a clearer and more detailed molecular theoretical basis for the prevention and control of stripe rust.【Method】Wheat varieties Shuiyuan 11 and stripe rust pathotypes CYR23 and CYR31 were used as experimental materials.Real-time quantitative PCR,tobacco subcellular localization assay,and virus-induced gene silencing technology (BSMV-VIGS) were used to study how the TaRING2 gene functioned in the interaction between wheat and stripe rust.【Result】The TaRING2 gene was 1 473 bp in length,encoding 490 amino acids,with a relative molecular weight of about 51.8 ku and an isoelectric point of 9.15.The qRT-PCR analysis results showed that compared with the affinity system of wheat and stripe rust,TaRING2 was extremely significantly upregulated in the nonaffinity system of wheat and stripe rust interaction at 12 and 24 h after vaccination,with a relative expression level about 10 times higher than that of the affinity system of wheat and stripe rust.The results showed that wheat plants for which the TaRING2 gene was silenced had reduced resistance to stripe rust,resulting in an increase in its spore production.Histological observations showed that in wheat plants where the TaRING2 gene was silenced,the hyphal development rate of stripe rust fungus was accelerated.【Conclusion】The E3 ubiquitin ligase TaRING2 gene regulates the resistance of stripe rust in wheat.
