不同赤霉素和浸泡时长对茶树嫩枝长穗扦插生根的影响

摘要:
【目的】研究生长调节剂中添加赤霉素和浸泡不同时长对茶树嫩枝长穗扦插生根的影响,为无性系茶树良种育苗提供参考。【方法】以5年生中茶108当年生嫩枝为材料,在生长调节剂浸泡液(100.00 mg/L NAA+50.00 mg/L IBA)中加入不同质量浓度(0,0.5,1.0,5.0,10.0,20.0和50.0 mg/L)赤霉素(GA3和GA7),并设浸泡不同时长(10~18 h)处理,通过统计扦插苗成活率、生根类型和生根率,以及根长、根直径、根体积、根表面积、根投影面积和根尖数等生根指标数据,研究GAs和浸泡时长对嫩枝长穗扦插生根的影响。【结果】所有处理茶树嫩枝扦插成活率为100%。在浸泡液中加入0.5 mg/L GA3培养9周后,生根率为87.50%,扦插苗生根指标除根尖数外,在所有GA3处理中最优;在浸泡液中加入50.0 mg/L GA7培养5周后生根率可达84.38%,培养9周后生根率为93.75%,扦插苗生根指标除根直径、根尖数外,在所有GAs处理中最优;其他GAs处理扦插苗生根率和各生根指标均优于对照,生根类型前期以皮部生根为主,随时间延长向混合生根过渡。扦插苗在生长调节剂浸泡液中浸泡11,12 h后,各生根指标总体最优,浸泡12 h时生根率最高,生根类型以愈伤生根为主,少量混合生根。【结论】在含100.00 mg/L NAA+50.00 mg/L IBA的生长调节剂浸泡液中添加50.00 mg/L GA7,并将茶树嫩枝浸泡12 h可有效提升其扦插生根效果;考虑到成本,赤霉素也可用0.5 mg/L GA3替代。

Effect of different gibberellins and soaking time on rooting of softwood long cuttings of tea plant

Abstract:
【Objective】Effects of adding gibberellins to the growth regulators and different soaking time on the rooting of softwood long cuttings of tea plant were studied,to provide a reference for the breeding of improved varieties of asexual tea plant.【Method】Long cuttings of 5-year-old Camellia sinensis (L.) O.Kuntze cv.Zhongcha 108 were used to investigate the effects of the growth regulator(100.00 mg/L NAA+50.00 mg/L IBA) added with gibberellins (GA3/GA7) of different mass concentration(0,0.5,1.0,5.0,10.0,20.0 and 50.0 mg/L)for different time duration(10-18 h).The effects of GAs and soaking time on the rooting of softwood long cuttings of tea plant were studied by collecting the rooting index data (the root survival rate,rooting types,rooting rate,root length,root diameter,root volume,root surface,root projected area and the number of root tips).【Result】The survival rate of cuttings was 100%.The rooting rate was 87.50% after 9 weeks of soaking treatment added with 0.5 mg/L GA3,and its rooting indicators of cuttings were the best among GA3 treatments,except the root tip number.The rooting rate reached 84.38% after 5 weeks of soaking treatment added with 50.0 mg/L GA7,and reached 93.75% rooting rate after 9 weeks of soaking treatment added with 50.0 mg/L GA7,showing the best rooting effect.All the rooting rate and rooting indicators of the GAs treatments were better than the control.The rooting type was mainly epidermal rooting in the early stage of rooting and was transitioned to mixed rooting with time.The best rooting indicators were observed in soaking treatment of growth regulator solution of 11 and 12 hours,with the highest rooting rate observed in treatment of 12 hours.The rooting type was mainly callus rooting with small amount of mixed rooting.【Conclusion】Soaking treatment of growth regulator soaking solution of 100.00 mg/L NAA+50.00 mg/L IBA added with 50.00 mg/L GA7 for 12 h could effectively promote the rooting of tea cuttings.To reduce the cost,gibberellin can also be replaced by 0.5 mg/L GA3.
