桉树枝瘿姬小蜂与寄主及寄生蜂体内细菌相似性研究

摘要:
【目的】研究桉树枝瘿姬小蜂、寄主桉树及其天敌孟氏胯姬小蜂三者体内细菌的相似性,明确内生细菌在桉树枝瘿姬小蜂入侵过程中的作用,为桉树枝瘿姬小蜂入侵和爆发机制研究提供参考。【方法】以巨园桉DH201-2叶柄和叶脉,无羽化孔虫瘿组织,南宁桉树枝瘿姬小蜂幼虫和雌成虫,四川桉树枝瘿姬小蜂雌成虫和孟氏胯姬小蜂成虫为材料,利用Illumina HiSeq 2500平台进行高通量测序方法,对桉树枝瘿姬小蜂、寄主桉树及其天敌孟氏胯姬小蜂三者体内细菌的多样性进行比较分析。【结果】在桉树枝瘿姬小蜂、寄主桉树及其天敌孟氏胯姬小蜂三者体内共发现24个门,55个纲,126个目,225个科和497个属的细菌。Alpha多样性分析表明,巨园桉DH201-2和无羽化孔虫瘿组织体内细菌的丰富度和多样性最高;桉树枝瘿姬小蜂幼虫和成虫体内菌群具有一定的相似性;桉树枝瘿姬小蜂成虫体内菌群组成与孟氏胯姬小蜂成虫体内菌群组成相似。变形菌门(Proteobacteria)是所有样本的优势菌门;甲基杆菌属(Methylbacillus)为巨园桉DH201-2的优势菌属,链球菌属(Streptococcus)为无羽化孔虫瘿组织内生菌的优势菌属,立克次氏体(Rickettsia)为桉树枝瘿姬小蜂幼虫,成虫和孟氏胯姬小蜂成虫内生菌的优势菌属。【结论】桉树、桉树枝瘿姬小蜂和孟氏胯姬小蜂三者体内细菌具有一定相似性,表明三者在共有细菌之间具有密切联系。

Study on the similarity of endosymbiotic bacteria among Leptocybe invasa and its host and parasite

Abstract:
【Objective】This study investigated the similarity of endosymbiotic bacteria among eucalyptus,Leptocybe invasa and Quadrastichus mendeli and clarified the role of endosymbiotic bacteria in the invasion of L.invasa to provide basis for understanding invasion and outbreak mechanism of L.invasa.【Method】The petiole and vein of DH201-2 (E.grandis×E.tereticornis),tissue without emergence holes gall,larvae and female L.invasa from Nanning,as well as Q.mendeli and female L.invasa from Sichuan were collected for analysis.The endosymbiotic bacteria of eucalyptus,L.invasa and Q.mendeli were analyzed and compared using Illumina HiSeq 2500.【Result】A total of 24 phyla,55 classes,126 orders,225 families and 497 genera were detected from eucalyptus,L.invasa and Q.mendeli.The Alpha diversity showed that the abundance and diversity of bacteria in eucalyptus DH201-2 and gall tissues were the highest.The endosymbiotic bacteria of L.invasa adults and larvae had certain similarity and the endosymbiotic bacteria of L.invasa and Q.mendeli was also similar.Proteobacteria was the dominant phylum in all samples,Methylbacillus was the dominant genus in DH201-2,Streptococcus was the dominant genus in gall tissues,and Rickettsia was the dominant genera in L.invasa larvae,adult and Q.mendeli.【Conclusion】The endosymbiotic bacteria had certain similarity among eucalyptus,L.invasa and Q.mendeli,indicating close relationship in endosymbiotic bacteria among them.
