表皮生长因子对21日龄肉鸡肠道菌群多样性的影响

摘要:
【目的】研究表皮生长因子(EGF)对肉鸡肠道菌群的影响。【方法】将72只1日龄AA肉鸡随机分成对照组(CK)、K组、E组和EK组,每组3个重复,每个重复6只鸡。CK组肉鸡饲喂基础饲粮,K组肉鸡饲喂添加250 mg/kg盐酸金霉素的饲料,E组肉鸡饲喂添加500 mg/kg EGF的饲料,EK组肉鸡饲喂添加500 mg/kg EGF和250 mg/kg盐酸金霉素的饲料。饲喂期21 d。21日龄时采集肉鸡盲肠内容物样本,采用16S rDNA基因分析方法对肉鸡盲肠中的细菌群落进行评价,利用 QIIME等测序分析软件对肉鸡盲肠中的菌群进行分类。【结果】1)菌种组成结果表明,无论是OTUs总数还是独有的OTUs数量,E组的菌种数量较CK组、K组高,而与EK组的数量接近。2)优势菌群分布结果表明,在门水平上,厚壁菌门在E组中含量最高,而K组与CK组中变形菌门水平相当;在种水平上,与CK组相比,E组肉鸡肠道中乳酸菌数量显著升高。3)Alpha 多样性分析表明,E组中观察到的菌种数、菌种多样性指数和菌种丰富度指数高于CK组和K组,但无显著性差异(P>0.05)。4)功能预测结果表明,CK组肠道微生物在遗传信息处理方面的功能较丰富,EK组在有机体系统、代谢作用和细胞工程方面的功能较丰富,E组各类功能较为均衡,而K组除未分类功能外,主要以环境信息处理功能较为丰富。【结论】喂饲500 mg/kg的EGF有助于提高肉鸡肠道内菌群多样性,并能改善肠道菌群结构,提高以乳酸菌为代表的厚壁菌门菌群丰度,降低以大肠杆菌为代表的变形菌门菌群含量。

Effect of epidermal growth factor on diversity of intestinal microflora in 21-day-old broilers

Abstract:
【Objective】This study investigated the effect of epidermal growth factor (EGF) on intestinal flora in broilers at different ages.【Method】A total of 72 1-day-old AA broilers were randomly divided into control group (CK),K group,E group and EK group,with 3 repeats in each group and 6 replicates in each repeat.Broilers in group CK were fed on basal diet,broilers in group K were fed on 250 mg/kg chlortetracycline hydrochloride,broilers in group E were fed on 500 mg/kg EGF,and broilers in EK group were fed on 500 mg/kg EGF test product and 250 mg/kg chlortetracycline hydrochloride.The trial period lasted for 21 days.A sample of 21-day-old broiler feces was collected,16S rRNA gene analysis method was used to evaluate and analyze the bacterial community in the cecum of broiler chickens,and sequencing analysis software such as QIIME was used to classify and analyze the bacterial community in broiler feces.【Result】1) Species composition showed that whether it was the total number of OTUs or the number of unique OTUs,the number of strains in the E group was higher than that in the CK and K groups,but was close to the number in the EK group.2) The distribution of dominant species showed that Firmicutes had the highest content in E group at the phylum level,while K and CK groups had the same level of Proteobacteria.At the species level,the number of lactic acid bacteria in the intestines of broilers in E group increased in comparison with CK.3) Alpha diversity analysis showed that number of species,species diversity index and species richness index in E group were higher than those in CK and K groups without significant differences (P>0.05).4) The function prediction results showed that gut microbes in CK group had richer functions in genetic information engineering,EK group had richer functions in organic systems,metabolism and cell engineering,E group had more balanced functions,and K group had more abundant environmental information processing functions.【Conclusion】Feeding 500 mg/kg EGF increased diversity and improved structure of intestinal flora in broilers.It also increased species abundance of Firmicutes represented by lactic acid bacteria and reduced contents of Proteobacteria species represented by Escherichia coli.
