2株林麝肺源铜绿假单胞菌毒力及致病性比较

摘要:
【目的】比较表型差异的林麝肺源铜绿假单胞菌ZP-1株(不产绿脓菌素)和BX-1株(产绿脓菌素)的毒力表达情况以及在小鼠急、慢性肺部感染时的致病性,为林麝化脓性肺炎的治疗与防控提供依据。【方法】比较ZP-1株和BX-1株的毒力因子表达差异,利用无创式气管插管滴注和制备琼脂糖包埋株的方法,建立ZP 1和BX 1株的小鼠急、慢性肺部感染模型,并检测小鼠急性、慢性感染肺脏残留铜绿假单胞菌数、白细胞总数和细胞因子含量,观察病理组织学变化。【结果】在体外培养条件下,ZP-1株的脓青素和胞外多糖表达量显著高于BX-1株(P<0.05);BX-1株的胞外蛋白酶(LasA、LasB)活性高于ZP-1株,但二者差异不显著(P>0.05);BX-1株鼠李糖脂表达量显著高于ZP-1株(P<0.05);ZP-1株与BX-1株的脂多糖结构具有明显差异,特别是核心多糖处。小鼠急性肺部感染12 h时,ZP-1组小鼠肺组织铜绿假单胞菌残留数、白细胞总数和细胞因子水平均高于BX-1组小鼠;感染24 h时,BX-1组小鼠IL-1β、IL-6含量超过ZP-1组。随着慢性肺部感染时间的延长,ZP-1组和BX-1组小鼠肺组织铜绿假单胞菌残留数、白细胞总数和IFN-γ含量均逐渐降低,IL-1β和IL-6含量均逐渐升高;除感染后第3天外,其余感染时间段ZP-1组小鼠肺组织白细胞总数均高于BX-1组;除感染后第14天外,其余感染时间段ZP-1组小鼠肺组织IL-1β、IL-6含量均高于BX-1组;在同一感染时期ZP-1组和BX-1组小鼠肺组织IFN-γ含量无明显差异。【结论】林麝肺源铜绿假单胞菌ZP-1株对肺部感染的小鼠致病性更强。

Comparison of virulence and pathogenicity of two Pseudomonas aeruginosa strains from lung of forest musk deer

Abstract:
【Objective】This study compared virulence expression of two Pseudomonas aeruginosa strains of ZP-1 (non producing pyocyanin) and BX-1 (producing pyocyanin) from lungs of forest musk deer and their pathogenicity in acute and chronic lung infections in mice to provide basis for treatment,prevention and control of purulent pneumonia in forest musk deer.【Method】The differences in virulence expression of ZP-1 and BX-1 strains were compared and mice acute and chronic lung infection models were established using non-invasive tracheal intubation and preparation of chronic infection-embedded strains.The number of residual Pseudomonas aeruginosa,total number of white blood cells,and level of cytokines in mice lungs with acute and chronic infections were detected and histopathological changes were observed.【Result】Expression of pyoverdine and extracellular polysaccharide of ZP-1 in vitro was significantly higher than that of BX-1 (P<0.05).The extracellular proteases (LasA and LasB) activities of BX-1 strain were higher than those of ZP-1 without significant differences (P>0.05).The expression of rhamnolipid in BX-1 was significantly higher than that in ZP-1 (P<0.05).The structure of lipopolysaccharides between ZP-1 and BX-1 was different,especially for core polysaccharides.At 12 hours of acute lung infection in mice,the residual number of Pseudomonas aeruginosa,total number of white blood cells and cytokine levels in lung tissues of the ZP-1 group were higher than those of the BX-1 group.At 24 h of infection,the levels of IL-1β and IL-6 in the BX-1 group were higher than those in the ZP-1 group.With the prolongation of chronic lung infection,the residual number of Pseudomonas aeruginosa,total number of white blood cells and content of IFN-γ in lung tissues of the ZP-1 and BX-1 groups gradually decreased,while contents of IL-1β and IL-6 gradually increased.The total number of white blood cells in the ZP-1 group was higher than that in the BX-1 group except for the third days after infection.The levels of IL-1β and IL-6 in lung tissues of the ZP-1 group were higher than those in the BX-1 group except for the 14th day after infection.There was no significant difference in IFN-γ content in lung tissues among the two strains.【Conclusion】The ZP-1 strain of Pseudomonas aeruginosa from lung of forest musk deer was more pathogenic to mice with lung infection.
