芜菁膏超声提取工艺优化及其抗氧化活性研究

摘要:
【目的】优化芜菁膏的超声法制备工艺并分析其抗氧化活性,为芜菁膏的高效制备和利用提供支持。【方法】以芜菁膏的出膏率为评价指标,通过单因素试验研究超声功率(400,500,600,700,800 W)、超声时间(40,50,60,70,80 min)、料(g)液(mL)比(1∶6,1∶8,1∶10,1∶12,1∶14)、提取次数(1,2,3,4,5次)对出膏率的影响。用Plackett-Burman L12(24)正交试验筛选出对出膏率影响最显著的3个因素,通过三因素三水平的响应面法优化芜菁膏的制备工艺。通过DPPH、ABST+自由基清除试验和细胞存活率试验评估芜菁膏的抗氧化活性。【结果】单因素试验得到芜菁膏的制备条件为:超声功率为600 W,提取时间为60 min,料液比为1∶10,提取次数为2次。根据Plackett Burman法和响应面法优化试验,得到芜菁膏的最佳制备工艺条件为:超声功率为614 W,提取时间为63 min,料液比为1∶9.5,提取次数为2次,在此条件下出膏率为59.02%。抗氧化活性试验结果表明,芜菁膏对DPPH和ABST+自由基的半数清除率分别为277.54和381.26 μg/mL,同时能够改善H2O2诱导的Raw 264.7细胞氧化应激损伤,提高细胞的存活率至82.57%。【结论】获得了超声法制备芜菁膏的最佳工艺条件;芜菁膏具有一定的抗氧化活性。

Optimization of ultrasonic extraction technology of Brassicaradix and its antioxidant activity

Abstract:
【Objective】In this experiment,the optimal preparation conditions and antioxidant activity of Brassica radix were studied to provide support for its efficient preparation and utilization.【Method】Single factor experiments were used to explore influences of ultrasonic power (400,500,600,700 and 800 W),ultrasonic time (40,50,60,70 and 80 min),material (g) to liquid (mL) ratio (1∶6,1∶8,1∶10,1∶12 and 1∶14) and extraction times (1,2,3,4 and 5) on yield rate, and the top three factors were selected by Plackett-Burman L12(24)experiment.Then,the preparation process of Brassica radix was optimized by the response surface method.The antioxidant activity of Brassica radix was evaluated by DPPH and ABST+ free radicals scavenging tests,as well as cell viability experiment.【Result】Through single factor experiments,the optimal preparation conditions of Brassica radix included ultrasonic power of 600 W,extraction time of 60 min,material=liquid ratio of 1∶10,and extraction times of 2.According to the Plackett-Burman and response surface method,the optimal process conditions included ultrasonic power of 614 W,extraction time of 63 min,material-liquid ratio of 1∶9.5,and number of extraction times of 2.Under these conditions,the yield of Brassica radix extract was 59.02%.Anti-oxidation tests showed that the half elimination ratios for DPPH and ABST+ free radical scavenging were 277.54 and 381.26 μg/mL,respectively.Brassica radix alleviated the oxidative stress damage of cells induced by H2O2 and increased survival rate of Raw 264.7 cells to 82.57%.【Conclusion】This study obtained the best conditions for preparation of Brassica radix by ultrasonic method.The Brassica radix exhibited certain antioxidant activity.
