不同果型黄瓜子房的组织学观察及其与内源激素含量的关系

摘要:
【目的】探明黄瓜果型与子房细胞数目、细胞大小及内源激素含量的关系,在组织学和激素水平上揭示黄瓜果型差异的机制。【方法】以12个不同果型黄瓜为试材,于各自开花当天采集子房,测量其长度、横径、种腔横径,计算子房形状指数,并采用石蜡切片技术观察子房纵切面和横切面的细胞数目、面积和密度,利用液质联用仪LC-MS/MS测定子房中内源激素生长素(IAA)、玉米素(ZT)、反式玉米素核苷(tZR)、赤霉素(GA3)、脱落酸(ABA)和茉莉酸(JA)的含量,分析子房大小及形状、细胞数目及面积和内源激素含量间的相关性。【结果】不同果型黄瓜开花当天的子房大小及形状、细胞数目及面积和内源激素含量不同,一些自交系间存在显著差异。子房形状指数与子房长度极显著正相关,与子房横径显著负相关;子房纵切面细胞数目与子房长度和子房形状指数极显著正相关,子房横切面细胞数目与子房横径和子房形状指数分别极显著正相关和显著负相关。IAA含量与子房长度和子房形状指数极显著正相关,与子房横径极显著负相关;ZT含量与子房横径显著正相关,tZR含量与子房横径显著负相关,GA3含量与子房长度和子房形状指数极显著负相关。【结论】不同果型黄瓜子房大小及形状的差异主要由细胞数目不同所致,IAA、ZT、tZR、GA3的含量均与子房大小及形状有关。

Histological observation and endogenous hormone content for cucumber ovaries with different fruit sizes and shapes

Abstract:
【Objective】This study explored the relationship between cucumber fruit size and shape with ovary cell number and size as well as endogenous hormone contents and revealed the mechanism for differences in cucumber fruit size and shape at histology and hormone levels.【Method】A total of 12 cucumbers with different fruit sizes and shapes were selected and ovaries at anthesis were collected to measure length,diameter and seed cavity diameter and calculate ovary shape index.Cell number,area,and density of the longitudinal and cross sections of ovaries were observed through paraffin section.The contents of endogenous hormones including IAA,ZT,tZR,GA3,ABA and JA for cucumber ovaries were determined by LC-MS/MS.The correlations between ovary size and shape traits,cell number and area and endogenous hormone contents were also analyzed.【Result】The ovary size and shape,cell number and area and endogenous hormone contents of ovaries with different fruit sizes and shapes were different,and there were significant differences among some inbred lines.The ovary shape index was very significantly positively correlated with ovary length and negatively correlated with ovary diameter.The ovary cell number in the longitudinal section had highly and significantly positive correlations with both ovary length and ovary shape index.The ovary cell number in the cross section showed significantly positive and negative correlations with ovary diameter and ovary shape index.The IAA content was very positively correlated with ovary length and ovary shape index,while negatively correlations were observed between ovary diameter and IAA content.The ovary diameter had significantly positive and negative correlations with ZT and tZR contents.GA3 content showed a strong significant negative correlation with both ovary length and ovary shape index.【Conclusion】The variations in ovary size and shape among cucumbers with different fruit sizes and shapes were mainly caused by differences in cell number.The contents of endogenous IAA,ZT,tZR and GA3 were all related to ovary size and shape.
