POU1F1基因多态性与藏羊生长性状的关联性分析

摘要:
【目的】探讨POU1F1基因的多态性与藏羊生长性状之间的关联性,寻找与藏羊生长性状相关的分子标记。【方法】利用DNA测序方法检测藏羊POU1F1基因的多态位点并对其进行基因分型,分析单一多态位点不同基因型及双倍型与藏羊生长性状的关联性。【结果】在POU1F1基因第4内含子上检测出1个多态位点g.14205G>A,在第5内含子上检测出2个多态位点g.15223G>A和g.15286A>G。关联性分析表明,g.14205G>A位点上AA基因型的体质量和体长分别极显著(P<0.01)和显著(P<0.05)高于GG和AG基因型;g.15223G>A位点上GG基因型的体质量显著高于GA基因型(P<0.05),极显著高于AA基因型(P<0.01);g.15286A>G位点AA基因型体质量极显著高于GG基因型(P<0.01),体高显著高于GG基因型(P<0.05)。单倍型Hap7(-GGA-)的发生频率最高,达到42.40%,其次为单倍型Hap5(-GAA-)和单倍型Hap3(-AGA-),发生频率分别为21.50%和14.70%。此外,POU1F1r2值均小于0.33,说明这3个多态位点之间不存在强的连锁性。通过合并基因型发现,双倍型H7H7(GGA-GGA)的各个生长性状表现最优。【结论】POU1F1基因的3个SNP位点可用于藏羊生长性状的选育。

Association of POU1F1 gene polymorphism with growth traits of Tibetan sheep

Abstract:
【Objective】The aim of this study was to investigate the association of single nucleotide polymorphism (SNP) of POU1F1 gene with Tibetan sheep growth traits,and to search for molecular markers related with growth traits.【Method】DNA sequencing method was applied to detect and genotype the polymorphism of POU1F1 gene in Tibetan sheep,and the association of polymorphism and combined genotypes with growth traits of Tibetan sheep was analyzed.【Result】One SNP of g.14205G>A was found in POU1F1 gene intron 4,and two SNPs of g.15223G>A and g.15286A>G were found on intron 5.Association analysis showed that AA individuals exhibited extremely larger body weight (P<0.01) and body length (P<0.05) than GG and AG individuals at g.14205G>A.The body weights of GG individuals were also significantly larger (P<0.01) than those with AA at g.15223G>A.AA individuals also demonstrated extremely larger body weight (P<0.01) and significantly larger body height (P<0.05) than GG at g.15286A>G.Hap7 (-GGA-) had the highest haplotype frequency (42.40%),followed by Hap5 (-GAA-),and Hap3 (-AGA-) with frequencies of 21.50% and 14.70%,respectively.The r2 values in POU1F1 were lower than 0.33,indicating that the three SNPs had no strong linkage disequilibrium (LD).The optical combined genotype was diplotype H7H7 (GGA-GGA),which was more effective on growth traits than other combined genotypes.【Conclusion】Three single nucleotide polymorphisms (SNPs) of POU1F1 gene could be used as candidate molecular markers for growth traits selection of Tibetan sheep.
