小麦根际产ACC脱氨酶细菌种群的多样性研究

摘要:
【目的】对杨凌和长武两地小麦根际土壤中能产生1-氨基环丙烷-1-羧酸(ACC)细菌的多样性进行调查比较。【方法】以ACC为唯一氮源从小麦根际土壤中富集和纯化产ACC脱氨酶细菌,使用Mi-seq高通量测序平台对富集产物进行多样性检测,构建16S rDNA基因克隆文库,对所分离的产ACC脱氨酶细菌进行系统发育分析;利用R语言绘图,对两地区该类细菌群落结构进行对比研究。【结果】长武地区小麦根际土壤2个样品中优化后的序列共计55 715条,基于97%的相似性被划分为35个OTU,Shannon多样性指数为1.43和0.6,Simpson多样性指数为0.380 5和0.773 7,经系统发育分析,主要隶属于假单胞菌属(Pseudomonas)、肠杆菌属(Enterobacteriaceae)、无色杆菌属(Achromobacter)、不动杆菌属(Acinetobacter)和乳球菌属(Lactococcus),所占比例分别为73.26%,15.86%,4.03%,3.26%和1.87%。杨凌地区小麦根际土壤2个样品中优化后序列共计34 044条,基于97%的相似性被划分为37个OTU,Shannon多样性指数为2.21和2.38,Simpson多样性指数为0.181 7和0.140 4,经系统发育分析,主要隶属于肠杆菌属(Enterobacteriaceae)、无色杆菌属(Achromobacter)、假单胞菌属(Pseudomonas)、芽孢杆菌属(Bacillus)、寡养单胞菌属(Stenotrophomonas),所占比例分别为37.66%,23.51%,14.90%,9.07%和5.80%。【结论】杨凌、长武地区小麦产ACC脱氨酶根际促生细菌的种类有部分重叠,但丰度差异较大,在空间分布上呈现一定程度的地缘隔离,调查结果可用于指导产ACC脱氨酶根际促生菌的资源开发及施用效果研究。

Abstract:
【Objective】This study investigated and compared the community diversity of bacteria producing ACC deaminase in rhizosphere of wheat in Yangling and Changwu.【Method】The bacteria producing ACC deaminase was enriched and purified using ACC as the sole source of nitrogen.The diversity was detected with Mi-seq high throughput sequencing platform.The bacterial community structure was analyzed by R language graph after constructing the clone library of 16S rDNA and phylogenetic analysis. 【Result】From the 2 Changwu samples,55 715 sequences were optimized and divided into 35 OTUs basing on 97% similarity.The Shannon indexes were 1.43 and 0.6,and the Simpson’s diversity indexes were 0.380 5 and 0.773 7.Pseudomonas (73.26%),Enterobacteriaceae (15.86%),Achromobacter (4.03%),Acinetobacter (3.26%) and Lactococcus (1.87%) were the dominate genera based on phylogenetic analysis.From the 2 Yangling samples,34 044 sequences were optimized and divided into 37 OTUs basing on 97% similarity.The Shannon indexes were 2.21 and 2.38,and the Simpson’s diversity indexes were 0.181 7 and 0.140 4.Enterobacteriaceae (37.66%),Achromobacter (23.51%),Pseudomonas (14.90%),Bacillus (9.07%) and Stenotrophomonas (5.80%) were the dominate genera according to phylogenetic analysis. 【Conclusion】There were more than 62.96% overlapping species of bacteria producing ACC deaminase in rhizosphere of wheat in Yangling and Changwu but there were large difference in abundance.There was geographical isolation in spatial distribution.
