Ghrelin对绵羊早期体外胚胎发育调节机制的初步研究

摘要:
【目的】探讨Ghrelin对绵羊早期胚胎发育的调节机制。【方法】将绵羊体外受精早期胚胎在Ghrelin组(300,1 000,10 000 ng/mL)、Ghrelin受体拮抗剂D-Lys3-GHRP-6组(10,200,400 ng/mL)、D-Lys3-GHRP-6+Ghrelin组(D-Lys3-GHRP-6 400 ng/mL,Ghrelin 300,1 000和2 000 ng/mL)、空白对照(不加D-Lys3-GHRP-6和Ghrelin)4个组别的胚胎发育液中进行发育培养,采用荧光定量PCR技术分别对各处理不同发育阶段(2-细胞期、4-细胞期、8-细胞期和16-细胞期)绵羊早期胚胎中ERK1、ERK2、P90rskBcl-2、Bax mRNA的表达情况进行检测。【结果】与空白对照组相比,在添加Ghrelin组ERK1、ERK2、P90rskBcl-2基因mRNA的表达表现为显著性上调(P<0.05),且这种上调作用可被D-Lys3-GHRP-6所拮抗;在添加Ghrelin组Bax基因mRNA的表达无显著性变化,但在D-Lys3-GHRP-6组Bax基因mRNA的表达表现为显著性上调(P<0.05)。【结论】Ghrelin可级联激活MAPK信号通路,抑制促凋亡基因Bax的表达、促进抗凋亡基因Bcl-2的表达,进而促进绵羊早期胚胎的发育。

Regulation of early ovine embryos development in vitro by Ghrelin

Abstract:
【Objective】The purpose of this study was to investigate the regulation mechanism of ghrelin during the development of early ovine embryos.【Method】Early ovine embryos were cultured in four groups with different media:Ghrelin (300,1 000,and 10 000 ng/mL),D-Lys3-GHRP-6 (Ghrelin receptor antagonist;10,200,and 400 ng/mL),D-Lys3-GHRP-6 plus Ghrelin (D-Lys3-GHRP-6:400 ng/mL,Ghrelin 300,1 000,and 2 000 ng/mL),and the control group (D-Lys3-GHRP-6:0 ng/mL and Ghrelin 0 ng/mL).The mRNA expression levels of ERK1,ERK2,P90rsk,Bcl-2,and Bax were analyzed during the same stages( 2-cell,4-cell,8-cell,and 16-cell).【Result】Compared with the control group,mRNA expressions of ERK1,ERK2,P90rsk,and Bcl-2 were upregulated significantly (P<0.05),and the upregulated function could be blocked by D-Lys3-GHRP-6.Bax mRNA expression was insignificantly different in Ghrelin groups but it was upregulated significantly in D-Lys3-GHRP-6 group (P<0.05).【Conclusion】Ghrelin promoted the development of early ovine embryos by activing the MAPK pathway,inhibiting the expression of Bax gene and promoting the expression of Bcl-2 gene.
