共存体系中对虾优势腐败菌对副溶血弧菌生物被膜形成的影响

摘要:
【目的】评估对虾优势腐败菌对副溶血弧菌生物被膜形成的影响,为对虾生产中生物被膜的危害评估和控制提供理论支持。【方法】采用改良微孔板法,分别检测5种腐败菌羽状变形杆菌(Proteus penneri)camtE、camtG、camtJ,希瓦氏菌(Advenellasp)camtF和未命名菌(Unknown)camtB的胞体或其乙酸乙酯提取物,与3株副溶血弧菌(Vibrio parahaemolyticus,VP)ATCC33847、ATCC17802和CAMT13011共培养后生物被膜的形成量,并用光学显微镜和扫描电镜观察生物被膜的形成情况。【结果】羽状变形杆菌camtE、camtG、camtJ及希瓦氏菌camtF和未命名菌camtB的乙酸乙酯提取物对副溶血弧菌ATCC33847、ATCC17802、CAMT13011生物被膜形成的促进作用大于上述5种腐败菌胞体的促进作用,前者生物被膜洗脱液OD600相对值达到0.46~0.83,后者仅达到0.26~0.59。副溶血弧菌与5种腐败菌乙酸乙酯提取物共培养48 h后其生物被膜形成受到促进,洗脱液OD600相对值为0.47~0.84;其中羽状变形杆菌camtE、camtG和camtJ乙酸乙酯提取物对3株副溶血弧菌的促进作用最明显,其生物被膜洗脱液OD600均值分别为0.74,0.70和0.68。【结论】5株对虾优势腐败菌与副溶血弧菌共培养可使原本产膜能力较弱的副溶血弧菌形成较致密的生物被膜,而这种影响可能与菌群间的群体感应信号分子有关。

Effect of shrimp dominated spoilage organisms in co-culture system on biofilm formation of Vibrio parahaemolyticus

Abstract:
【Objective】This paper evaluated the biofilm formation of Vibrio parahaemolyticus organisms influenced by shrimp dominated spoilage bacteria.【Method】The biofilm formation of three strains of V.parahaemolyticus ATCC33847,ATCC17802 and CAMT13011 influenced by five spoilage bacterium of Proteus penneri camtE,camtG,camtJ,Advenellasp camtF and unknown camtB cells or ethyl acetate extracts was measured in vitro using a colorimetric 96-well microtitre plate.The biofilm production and morphology were also observed using optical microscope and scanning electron microscopy.【Result】Ethyl acetate extracts of Proteus penneri camtE,camtG,camtJ,and Advenellasp camtF and unknown camtB had stronger effects on biofilm development of ATCC33847,ATCC17802,and CAMT13011 than those cells,with a higher OD600 of 0.46-0.83 compared that of the cells (0.26-0.59).Biofilm formation of V.parahaemolyticus after 48 h co-culturing with 5 extracts was promoted with OD600 of 0.47-0.84.Ethyl acetate extracts of Proteus penneri camtE,camtG and camtJ played the most significant role in promoting biofilm production of 3 strains of V.parahaemolyticus with average OD600 of 0.74,0.70 and 0.68,respectively.【Conclusion】The biofilm development of V.parahaemolyticus was promoted by 5 specific spoilage organisms in co-culture system,which is likely related to quorum sensing among organisms.
