响应曲面法优化夏枯草中迷迭香酸的超声提取工艺

摘要:
【目的】采用响应曲面法建立Box Behnken模型,优化夏枯草中迷迭香酸的超声提取工艺。【方法】以料(g)液(mL)比(1∶10,1∶20,1∶30,1∶40,1∶50,1∶60)、乙醇体积分数(40%,50%,60%,70%,80%,90%)、提取时间(10,20,30,40,50,60 min)为考察因素,迷迭香酸提取率为考察指标,在单因素试验的基础上,采用3因素3水平响应曲面法分析,确定夏枯草中迷迭香酸的最优提取工艺。【结果】夏枯草中迷迭香酸最佳提取工艺参数为:料(g)液(mL)比1∶28,乙醇体积分数74%,提取时间55 min,在该工艺参数条件下,迷迭香酸提取率达0.52%,与预测值0.54%偏差较小,说明响应曲面法优化超声提取夏枯草中迷迭香酸工艺的参数准确可靠。【结论】得到了夏枯草中迷迭香酸提取的优化条件,本研究所用方法具有可行性和实用性。

Optimization of ultrasonic assisted extraction of rosmarinic acid from Prunella vulgaris by response surface method

Abstract:
【Objective】Ultrasonic assisted extraction of rosmarinic acid from Prunella vulgaris was optimized using response surface method and Box-Behnken model. 【Method】Solid liquid ratio (g/mL) (1∶10,1∶20,1∶30,1∶40,1∶50,and 1∶60),ethanol volume fraction (40%,50%,60%,70%,80%,and 90%),and extraction time (10,20,30,40,50,and 60 min) were tested to select optimal extraction conditions for rosmarinic acid from Prunella vulgaris.【Result】The optimum extraction parameters were:solid liquid ratio 1∶28 (g/mL),ethanol volume fraction 74%,extraction time 55 min,and the extraction rate was 0.52%,which was very close to the predicted value of 0.54%.The surface response optimization of rosmarinic acid ultrasonic extraction from Prunella vulgaris was accurate and reliable.【Conclusion】Optimal extraction conditions for rosmarinic acid from Prunella vulgaris were obtained and the method was feasible and practical.
