细胞周期蛋白家族基因在家蚕胚胎期的表达分析

摘要:
【目的】研究家蚕细胞周期蛋白家族基因(CyclinACyclinBCyclinB3、CyclinECyclinL1)在滞育卵与即时浸酸处理的滞育卵发育过程中的转录活性,为家蚕胚胎发育及细胞周期调控机制研究提供参考。【方法】以家蚕品种大造为试材,取产后1~8 d的滞育卵及产后20 h即时浸酸的非滞育卵,提取其RNA反转录成cDNA,采用实时荧光定量PCR方法分析家蚕Cyclin家族基因在胚胎发育过程中的转录水平。【结果】在即时浸酸处理的家蚕非滞育卵中均能检测到Cyclin家族基因的表达,但不同发育阶段Cyclin家族基因表达量差异很大,在发育开始的1~3 d,Cyclin家族基因变化显著,说明此期细胞分裂频繁,正处于组织和器官活跃形成时期。在胚胎发育晚期,除CyclinL1外的其他基因表达量都很低,表明此时幼虫器官发育几近完成,因此细胞分裂迟缓。而滞育卵从蛾体产下经过3 d后,细胞分化停滞于G2期,Cyclin家族基因也进入沉默状态。【结论】Cyclin家族基因在家蚕胚胎发育过程中具有重要的调控作用。

Expression of Cyclin family genes during embryonic stage of Bombyx mori

Abstract:
【Objective】The transcriptional activities of Cyclin family genes (CyclinA,CyclinB,CyclinB3,CyclinE,and CyclinL1) during development of diapause and non-diapause eggs of Bombyx mori were studied to provide reference for embryonic development and control mechanism of cell cycle in B.mori.【Method】The total RNA was isolated from 1-8 d diapause and 20 h non-diapause eggs of B.mori (Dazao) and was reversely transcribed into cDNA.The transcription levels of Cyclin family genes during embryonic development stage were measured using real-time fluorescence quantitative PCR.【Result】All of the Cyclin family genes were detected in non diapause eggs.The Cyclin family genes expression levels varied widely at different embryonic development stages.At 1-3 d,Cyclin family genes changed significantly,indicating that the cell division was frequent at this phase,and it was during the formation stage of tissues and organs.Expressions of all genes except CyclinL1 were low in late embryonic development stage,indicating that the larval organ development was almost complete at this time.Three days after the lay of diapause eggs,the differentiation stagnated at the G2 phase,and Cyclin family genes were in silenced state.【Conclusion】The Cyclin family genes play an important role in regulation at embryonic development stage.
