甘蓝型油菜萝卜质雄性不育恢复系R2572的分子标记研究

摘要:
【目的】对甘蓝型油菜萝卜质雄性不育系的18个恢复材料进行恢复基因Rfo及其旁侧特异性标记研究,以便筛选出与国外同类恢复材料具有差异的恢复系。【方法】以18个甘蓝型油菜萝卜质雄性不育恢复系为材料,选择1个Rfo特异性分子标记、5个与Rfo紧密连锁的标记以及先锋国际良种公司关于芸薹属Ogura恢复系专利鉴定的特异性标记对所选恢复材料进行分子检测,根据检测结果与国内外同类恢复材料(RRH1、R113、R211、R2000、CLR650、SRF系、NW1717)进行对比鉴定。【结果】18个萝卜质雄性不育恢复材料均含有恢复基因Rfo,各恢复系缺少了R113、R211,R2000共有的标记E33M47、E32M50、OPN20、OPH15、IN6RS4、E33M58、BolJon、ScH03,同时多了一个R211、R2000、CLR650共有的标记PGlint。各恢复系含有和NW1717相同的标记。【结论】R2572可能与RRH1、R113、R211、R2000、CLR650,SRF系是不同的,而与导入NW1717的萝卜片段长短较为接近。

Abstract:
【Objective】This study conducted molecular research on 18 Ogura CMS restorers (R2572) in Brassica napus with one specific marker for Ogura CMS restoring gene Rfo and its linkage markers to select different restorers from similar foreign materials.【Method】Molecular detection on 18 Ogura CMS restorers (R2572) was conducted using one Rfo specific marker,5 radish markers linked with Rfo,and specific markers of SRF line.Testing results were compared with similar restorers (RRH1,R113,R211,R2000,CLR650,SRF and NW1717).【Result】All the 18 restorers of R2572 contained Ogura CMS restorer gene Rfo.Compared to R113,R211 and R2000,R2572 lacked markers E33M47,E32M50,OPN20,OPH15,IN6RS4,E33M58,BolJon and ScH03 but owned PGlint.R2572 had the same markers as NW1717.【Conclusion】R2572 may be different from RRH1,R113,R211,R2000,CLR650 and SRF,and its length was similar to of radish fragment with NW1717.
