酸枣仁皂苷回流提取工艺的响应面优化

摘要:
【目的】采用中心组合试验 响应面法优选酸枣仁皂苷的最佳回流提取工艺,为酸枣仁皂苷提取的工业化生产提供依据。【方法】 采用HPLC UV检测酸枣仁皂苷A和B的含量,用乙醇回流提取法,以乙醇体积分数(0,30%,50%,70%,80%,95%)、液固比(8∶1,10∶1,12∶1,15∶1,20∶1)、提取时间(1,1.5,2,2.5,3 h)和提取次数(1,2,3,4)为考察因素,筛选各因素对酸枣仁皂苷提取影响的取值范围,在此基础上进行中心组合试验,用SAS(9.2)软件建立预测模型,响应面分析最佳提取条件。【结果】响应面分析优选得到的酸枣仁皂苷最佳提取工艺为:体积分数66%乙醇,液固比12.5∶1,回流提取3次,每次1.8 h。在此条件下,酸枣仁皂苷A和B含量之和平均为 1.21 mg/g,与模型预测值(1.30 mg/g)接近。【结论】用中心组合设计 响应面法优选的酸枣仁皂苷A和B提取工艺可信度高,皂苷含量验证值与模型预测值接近,模型预测效果好,工艺稳定,可用于指导酸枣仁皂苷的工业化提取生产。

Optimization of reflux extraction of jujuboside from Ziziphi spinosae semen by central composite design-response surface methodology

Abstract:
【Objective】 The reflux extraction of jujuboside from Ziziphi spinosae semen was optimized by central composite design-response surface methodology to provide basis for industrial extraction of jujuboside from Ziziphi spinosae semen.【Method】The contents of jujuboside A and B were detected by HPLC UV,using ethanol extraction.Different ethanol concentrations (0,30%,50%,70%,80%,and 95%),solvent-to-solid ratios (8∶1,10∶1,12∶1,15∶1,and 20∶1),extraction times (1,1.5,2,2.5,and 3 h) and extraction frequencies (1,2,3,and 4) were tested to screen value ranges.Then,central composite design (CCD) response surface methodology was used and a predictive model was developed by SAS(9.2) software to determine the optimum extraction conditions.【Result】The optimum reflux extraction conditions were:66% ethanol,solvent-to-solid ratio 12.5∶1,and extracted for 1.8 h and 3 times.The average content of jujuboside A and B was 1.21 mg/g,which was close to the model prediction (1.30 mg/g).【Conclusion】CCD-response surface methodology for optimizing the reflux extraction of jujuboside A and B was reliable,and the detected content of jujuboside was close to the predicted value. The predicted extraction process was also stabled and could be used to direct the industrialized extraction of jujuboside.
