产蛋前期和产蛋期鹅HPG轴OAZ1和OAZ2基因表达的研究

摘要:
【目的】探讨产蛋前期和产蛋期鹅HPG轴中OAZ1和OAZ2基因表达的差异。【方法】选取健康的产蛋前期和产蛋期四川白鹅母鹅各3只,采集下丘脑、垂体和卵巢组织样品,提取总RNA并反转录合成cDNA,应用实时荧光定量PCR检测产蛋前期和产蛋期四川白鹅下丘脑、垂体、卵巢组织中OAZ1和OAZ2基因的相对表达量。【结果】在产蛋前期和产蛋期四川白鹅下丘脑、垂体和卵巢组织中,OAZ1和OAZ2基因均有表达,并且产蛋期OAZ1和OAZ2表达量均极显著高于产蛋前期(P<0.01),产蛋期四川白鹅下丘脑、垂体和卵巢中OAZ1的表达量分别是产蛋前期的1.79倍(P<0.01)、2.96倍(P<0.01)和3.48倍(P<0.01);产蛋期四川白鹅下丘脑、垂体和卵巢中OAZ2的表达量分别是产蛋前期的2.66倍(P<0.01)、1.38倍(P<0.01)和2.03倍(P<0.01)。【结论】OAZ1和OOAZ2可在HPG轴的各级水平发挥其调控鹅繁殖过程的功能,OAZ1主要参与卵巢功能的调控,而OAZ2主要参与下丘脑功能的调控。

Expressions of OAZ1 and OAZ2 genes in HPG axis of goose at pre-laying and laying stages

Abstract:
【Objective】This study aimed to explore the differences in expressions of ornithine decarboxylase antizyme 1 (OAZ1) and ornithine decarboxylase antizyme 2 (OAZ2) genes in the hypothalamic-pituitary-gonadal axis of Sichuan white geese at pre-laying and laying stages.【Method】Healthy female Sichuan white geese at pre-laying and laying stages (n=3,respectively) were selected,and hypothalamic,pituitary and ovary tissues were collected.Total RNA was extracted and cDNA was synthesized.The expression levels of OAZ1 and OAZ2 in hypothalamus,pituitary gland and ovary samples were determined by quantitative real-time PCR.【Result】OAZ1 and OAZ2 genes expressed in hypothalamus,pituitary and ovarian tissues of Sichuan white goose at both pre-laying and laying stages,and the expressions of OAZ1 and OAZ2 at laying period were significantly higher than at pre-laying period (P<0.01).The expression levels of OAZ1 mRNA in hypothalamus,pituitary and ovarian tissues of geese at laying stage were 1.79 (P<0.01),2.96 (P<0.01) and 3.48 (P<0.01) times of those for geese at pre-laying stage,respectively.The levels of OAZ2 mRNA expression in hypothalamus,pituitary and ovarian tissue of geese at laying stage were 2.66 (P<0.01),1.38 (P<0.01) and 2.03 (P<0.01) times of those in geese at pre-laying stage,respectively.【Conclusion】OAZ1 and OAZ2 mediated the biological function of HPG axis.OAZ1 may play a more important role in ovary while OAZ2 may play a more important role in hypothalamic.
