脂联素基因在荷斯坦公牛不同组织部位的表达差异

摘要:
【目的】研究脂联素基因在荷斯坦公牛不同组织及月龄间的相对表达差异性,为脂联素基因作为荷斯坦公牛育肥指标的分子辅助标记提供依据。【方法】运用实时荧光定量PCR法检测了脂联素基因在15和17月龄荷斯坦公牛脂肪组织(背部脂肪、腰部脂肪、肾周脂肪、肠系膜脂肪)和肌肉组织(背最长肌和半腱肌)间的相对表达差异性。【结果】15月龄时脂联素基因在荷斯坦公牛肾周脂肪和肠系膜脂肪中的相对表达量极显著高于腰部脂肪、背部脂肪、背最长肌和半腱肌(P<0.01),17月龄时仅显著高于背最长肌和半腱肌(P<0.05);17月龄时该基因在背部脂肪中的相对表达量显著高于15月龄时(P<0.05)。【结论】脂联素基因在15和17月龄荷斯坦公牛脂肪组织和肌肉组织中均有表达,且在脂肪组织和肌肉组织间具有组织表达差异性。

Expression differences of ADIPOQ gene in different tissues of Holstein bull

Abstract:
【Objective】The objective of this study was to compare the expressions of adiponectin (ADIPOQ) gene in different tissues of Holstein bulls at different ages.【Method】Real-time fluorescent quantitative PCR was used to determine the relative expression of ADIPOQ gene using RNA isolated from different adipose tissues (subcutaneous fat over sirloin,subcutaneous fat over rib,kidney fat,and mesenteric fat) and muscle tissues (longissimus dorsi and semitendinosus) of Holstein bulls with two different ages (15 and 17 months).【Result】Expressions of ADIPOQ in kidney fat and mesenteric fat were greater (P<0.01) than in subcutaneous sirloin fat,subcutaneous rib fat,longissimus dorsi and semitendinosus in 15 month group and greater(P<0.05)than in longissimus dorsi and semitendinosus in 17 month group.Expressions of ADIPOQ gene also differed between 15 month and 17 month old Holstein bulls in the same adipose tissues.The expressions in subcutaneous rib fat in 17 month group were greater (P<0.05) than in 15 month group.【Conclusion】Expressions of ADIPOQ gene in different tissues of Holstein bulls with ages of 15 month and 17 month differed.These results suggested that evaluation of expressions of ADIPOQ gene should consider the age as well as the tissue.
