连翘苷对小鼠遗传物质的损伤作用

摘要:
【目的】评价连翘苷对小鼠遗传物质的损伤作用,为揭示连翘苷对动物细胞影响的机理及其临床运用提供参考。【方法】给小鼠腹腔注射1 500,1 250,1 000,750,500和250 mg/kg的连翘苷1次,连续观察10 d,记录小鼠死亡数,采用改良寇氏法计算连翘苷对小鼠的半数致死量(LD50)。然后给小鼠腹腔注射250,500,750和1 000 mg/kg的连翘苷连续5 d,取小鼠骨髓嗜多染红细胞、肝组织和小鼠精子样品,检测连翘苷对嗜多染红细胞微核率、肝细胞彗星试验细胞拖尾率和小鼠精子畸变率的影响。以腹腔注射羧甲基纤维素钠溶液和环磷酰胺的小鼠分别为阴性和阳性对照。【结果】连翘苷对小鼠的LD50为1 086 mg/kg。连翘苷剂量≥500 mg/kg时,嗜多染红细胞微核率和总核异常率极显著提高(P<0.01),1 000 mg/kg时微核率和总核异常率分别达到最高值2.28%和18.94%;在受试剂量下,彗星电泳肝细胞DNA拖尾率与阴性对照组相比,差异无统计学意义;与阴性对照组相比,连翘苷剂量≥500 mg/kg时小鼠精子畸形率显著或极显著提高。【结论】高剂量(≥500 mg/kg)连翘苷能使小鼠嗜多染红细胞微核率和雄性小鼠精子畸形率上升,有一定的遗传毒性;连翘苷对小鼠肝细胞DNA无损伤作用。

Damage of phillyrin to genetic material of mice

Abstract:
【Objective】This study evaluated the damage of phillyrin to genetic material of mice,aiming to reveal the mechanism how phillyrin could effect animal cell and to provide theoretical basis for its clinical application. 【Method】The median lethal dose (LD50) of phillyrin to mice was calculated using modified Karber’s method by intraperitoneal injecting 1 500,1 250,1 000,750,500 and 250 mg/kg phillyrin to mouse and counting the deaths for 10 d.The mice bone marrow polychromatic erythrocyte (PCE),hepatic tissue and sperm samples were collected to detect the effect of phillyrin on micronucleus (MN) frequency,hepatic cell comet tail rate and mouse sperm aberration rate by intraperitoneal injecting 250,500,750 and 1 000 mg/kg phillyrin to mice for 5 days.Injection of sodium carboxymethyl solution was used as negative control and injection of cyclophosphamide was used as positive control.【Result】The LD50 of phillyrin to mice was 1 086 mg/kg.MN frequency of PCE and the total nucleus abnormity rate in bone marrow increased very significantly (P<0.01) when phillyrin dose was ≥500 mg/kg.The highest frequency of MN and the total nucleus abnormity rate were 2.28% and 18.94% when phillyrin dose was 1 000 mg/kg.No statistical difference was found in comet tail DNA rate between phillyrin dose group and negative control group.Compared with negative control group,sperm aberration rate increased significantly (P<0.05) or very significantly (P<0.01) when phillyrin dose was ≥500 mg/kg.【Conclusion】Phillyrin had limited genotoxicity because high dose phillyrin (≥500 mg/kg) increased MN frequency of PCE and sperm aberration rate.However,phillyrin had no damage effect on DNA of hepatic cells in mice.
