2种乳酸杆菌肠道黏膜免疫调节作用的比较

摘要:
【目的】比较嗜酸乳酸杆菌(Lactobacillus acidophilus)和植物乳酸杆菌(Lactobacillus plantarum)肠道黏膜免疫调节作用的差异。【方法】以C57BL/6小鼠为受试动物,分别灌胃嗜酸乳酸杆菌和植物乳酸杆菌 7 d后取材,采用流式细胞术、ELISA等方法,对脾脏和肠系膜淋巴结中T细胞亚群、结肠PP结中TLR2+和TLR4+细胞、结肠组织中IFN-γ和IL-4水平及小肠sIgA水平进行检测,明确2种乳酸杆菌对肠道黏膜的免疫调节作用。【结果】2种乳酸杆菌对小鼠体质量无显著影响。2种乳酸杆菌均可显著提高小鼠肠系膜淋巴结中CD3+CD4+和CD3+CD8+T细胞比例,且与嗜酸乳酸杆菌相比,植物乳酸杆菌更倾向于提高CD3+CD8+T细胞比例;而与对照组相比,二者对脾脏中CD3+CD4+和CD3+CD8+T细胞比例均无显著影响。2种乳酸杆菌均可极显著提高结肠PP结中TLR2+细胞比例,而对TLR4+细胞比例无显著影响。2种乳酸杆菌均可极显著提高结肠组织中IFN-γ水平,嗜酸乳酸杆菌还可极显著提高IL-4水平。2种乳酸杆菌均能显著提高肠道黏膜sIgA水平。【结论】2种乳酸杆菌均可提高结肠PP结中TLR2+细胞比例,嗜酸乳酸杆菌上调Th1和Th2型免疫反应,植物乳酸杆菌上调Th1型免疫反应。

Comparison of the intestinal mucosal immunoregulation functions of two Lactobacillus strains

Abstract:
【Objective】This study compared the intestinal mucosal immunoregulation functions of Lactobacillus acidophilus and Lactobacillus plantarum.【Method】Seven days after intragastric administration with Lactobacillus acidophilus and Lactobacillus plantarum,C57BL/6 mice were killed,and T cells subgroups in spleen and MLNs,TLR2 and TLR4 positive cells in colon Peyer’s patch,IFN-γ and IL-4 concentration in colon tissues and sIgA level in small intestine were detected using Flow cytometry and ELISA.【Result】Both Lactobacillus strains had no significant effect in body weights of mice compared to the control.While both of them increased the ratios of CD3+CD4+ and CD3+CD8+T cells in colon Peyer’s patch,and Lactobacillus acidophilus had larger effect than Lactobacillus plantarum.However,there was no significant effect on ratios of CD3+CD4+ and CD3+CD8+T cells in spleen.Both strains could increase numbers of TLR2+ cells in colon Peyer’s patch but not for TLR4+ cells.Both Lactobacillus acidophilus and Lactobacillus plantarum increased IFN-γ production in colon tissue,and <Lactobacillus acidophilus also increased the level of IL-4.In addition,both Lactobacillus acidophilus and Lactobacillus plantarum could promote intestinal sIgA production.【Conclusion】Both strains increased TLR2 positive cells in the intestinal mucosal,and Lactobacillus acidophilus promoted Th1 and Th2 immune response whereas Lactobacillus plantarum enhanced Th1 immune response.
