5个美洲黑杨品种EST-SSR指纹图谱的构建及遗传差异分析

摘要:
【目的】利用新型分子标记EST-SSR,从分子水平上区分亲缘关系较近、形态上难以区分的5个美洲黑杨品种,并构建其指纹图谱。【方法】采用EST-SSR分子标记,从22对引物中筛选多态性引物,用以鉴别I-69杨、陕林3号、创新杨1号、南抗1号、北抗1号 5个美洲黑杨品种,并根据多态性条带计算各品种间的遗传相似系数,构建各自的指纹图谱。【结果】从22对引物中筛选出的17对多态性引物共扩增出86条谱带,其中多态性条带69条,多态性比率为80.23%,有5对引物可以将5个美洲黑杨品种完全区分开;各品种间的遗传相似系数为0.444 4~0.717 2,平均相似系数为0.580 8;构建了各品种的EST SSR特征指纹图谱。【结论】5个美洲黑杨品种间的遗传差异较小,其中创新1号与陕林3号遗传差异最大。

Construction of fingerprinting and analysis of genetic difference for 5 Populus deltoides varieties using EST-SSR markers

Abstract:
【Objective】Identification and fingerprinting construction of 5 Populus deltoides varieties with close genetic relationships and similar morphologies were conducted using novel EST-SSR markers.【Method】Polymorphic primers were selected from 22 pairs of EST-SSR markers to identify 5 Populus deltoides varieties,P.deltoides cv.‘Lux’(I-69/55),P.deltoides cl.‘SL-3’,P.deltoides cl.Xinshiji-l,P.deltoides cl.Nankang-1,and P.deltoides cl.Beikang-1(16-27/92),calculate their genetic similarity coefficients and construct their fingerprints with polymorphic bands.【Result】17 pairs of polymorphic primers were selected from 22 pairs of primers,and 86 bands were amplificated with 69 polymorphic bands.Polymorphic rate of the bands was 80.23%,and 5 primers could identify the 5 varieties completely.The similarity coefficients of Populus varieties ranged from 0.444 4 to 0.717 2 with a mean of 0.580 8.The EST-SSR unique fingerprint map for each variety was constructed.【Conclusion】The genetic differences among the studied 5 Populus deltoides varieties were small,and the largest genetic difference was between P.deltoides.cl.Xinshiji-l and P.deltoides cl.‘SL-3’.
