平欧杂交榛抗寒生理特性研究

摘要:
【目的】研究不同低温处理对平欧杂交榛枝条抗寒生理指标的影响,探讨杂交榛抗寒性与膜透性、渗透调节物质含量、膜脂过氧化性、过氧化物酶活性及萌芽率的关系,为杂交榛的高抗寒性育种和栽培提供依据。【方法】以平欧杂交榛品种新榛1号(84 254)、新榛2号(82-11)、新榛3号(84-310)和平顶黄(80-43)的1年生枝条为试验材料,分别于-15,-20,-25,-30,-35和-40 ℃的低温下处理24 h,以-15 ℃处理的枝条为对照,研究在低温条件下杂交榛枝条的电解质渗出率、游离脯氨酸(游离Pro)和丙二醛(MDA)含量、过氧化物酶(POD)活性及萌芽率的变化。【结果】在不同低温处理下,各榛子品种的电解质渗出率和MDA含量均随温度的降低而增高,其中新榛2号、平顶黄的电解质渗出率和MDA含量均高于新榛1号和新榛3号,2个指标以新榛1号最小;4个品种的游离Pro含量均随温度的降低而增高,以新榛1号最大,新榛3号次之,新榛2号和平顶黄较接近;POD活性和萌芽率均随温度的降低而降低,其中新榛1号始终保持最大。4个品种的上述生理指标、萌芽率与处理温度呈显著相关性;电解质渗出率与MDA含量、游离Pro含量、POD活性、萌芽率呈极显著相关性。同一品种的MDA含量在不同处理温度间差异达显著水平,而其他生理指标差异达极显著水平。【结论】综合分析各品种抗寒性生理指标,推测其抗寒性强弱顺序为:新榛1号最强,新榛3号次之,新榛2号较弱,平顶黄最弱;电解质渗出率、MDA含量、游离Pro含量、POD活性和萌芽率可以作为评价杂交榛抗寒性的生理指标。

Study on character of cold-resistance physiological of Corylus heterophylla×Corylus avellana 

Abstract:
【Objective】Effects of low temperature treatment on physiological indexes of hybrid hazel branches,and the relationship of hybrid hazel cold resistance and membrane permeability,content of osmotic adjustment material,membrane lipid peroxidation,peroxidase and germination rate,were investigated to improve breeding and cultivation planting of hybrid hazel varieties with cold resistance.【Method】1-year-old twigs of hybrid hazelnut varieties,Xinzhen 1(84-254),Xinzhen 2(82-11),Xinzhen 3(84 310),and Pingdinghuang (80-43) were pretreated for 24 hours with low temperatures of -15,-20,-25,-30,-35,and -40 ℃,respectively.Treatment with temperature of -15 ℃ was taken as the control.Changes of electrolyte leakage,contents of proline (Pro) and malondialdehyde (MDA),peroxidase (POD) activity and germination rate of branches were analyzed.【Result】Electrolyte leakage rate and MDA content of each hazelnut variety increased as the decrease of temperature.The electrolyte leakage rate and MDA content of Xinzhen 2 and Pingdinghuang were higher than those of Xinzhen 3,and those of Xinzhen 1 were the minimum.Pro contents of all four varieties increased along with the decrease of temperature,Xinzhen 1 had the maximum value,followed by Xinzhen 3,and contents of Xinzhen 2 and Pingdinghuang were similar.POD activity and germination rate decreased as the decrease of temperature,and those of Xinzhen 1 were the maximum.The physiological indexes and germination rate significantly correlated with the treatment temperature.Electrolyte leakage was significantly related to MDA content,Pro content,POD activity and germination rate.At different temperatures,MDA content of each variety was significant different,while other indexes were extremely significant different.【Conclusion】Based on comprehensive analysis of different physiological and biochemical indexes,the cold resistances of the varieties were in a decreasing order of:Xinzhen 1>Xinzhen 3>Xinzhen 2>Pingdinghuang.Electrolyte leakage,MDA,Pro,POD and germination rate could be used to evaluate cold-resistance of hazel varieties.
