超声波-双酶法协同提取玉米须黄酮工艺的优化

摘要:
【目的】采用响应面优化法,研究超声波-双酶法协同提取玉米须黄酮的最优工艺,为进一步开发玉米须资源提供依据。【方法】以黄酮提取率为指标,在超声波 双酶法协同提取玉米须黄酮单因素试验的基础上,采用Box-Behnken响应面设计,利用Design-Exper 7.0.0软件对玉米须黄酮提取率的二次回归模型进行分析,并对该工艺下的提取率与单一超声波法提取率进行比较。【结果】超声波-双酶法协同提取玉米须黄酮的最佳工艺参数为:液料比31∶1(mL/g)、超声功率173 W、酶解时间42 min、加酶比(果胶酶∶纤维素酶)1.9∶1,在此条件下黄酮提取率为(0.86±0.02)%,较单一超声波提取(提取率(0.72±0.02)%)有明显提高。【结论】用Box-Behnken 响应面法优化超声波-双酶法协同提取玉米须黄酮工艺是可行的。

Optimization of ultrasonic-double enzymatic synergistic extraction of flavonoids from corn silk

Abstract:
【Objective】The conditions for ultrasonic-double enzymatic synergistic extraction of flavonoids from corn silk by response surface methodology were optimized to improve the utilization of corn silk.【Method】On the basis of single-factor tests,Box-Behnken response surface design was used to optimize the ultrasonic-double enzymatic synergistic extraction of flavonoids from corn silk with a focus on extraction rate of flavonoids.Two regression mode of extraction rate of flavonoids was analyzed with Design Exper 7.0.0 software.The extraction rates of flavonoids of this method and single ultrasonic extraction were then compared as well.【Result】The optimal technological conditions for ultrasonic-double enzymatic synergistic extraction of flavonoids from corn silk were:liquid-to-material ratio was 31∶1(mL/g),ultrasonic power was 173 W,hydrolysis time was 42 min,and the ratio of double enzymatic (pectinase∶cellulose) was 1.9∶1.The extraction rate of flavonoids was (0.86±0.02)%,which was significantly higher than that of single ultrasonic extraction (0.72±0.02)%.【Conclusion】It is feasible to use Box-Behnken response surface method to optimize the ultrasonic-double enzymatic synergistic extraction of flavonoids from corn silk.
