2种水稻不同细胞质雄性不育的恢复基因分析

摘要:
【目的】由华南农业大学选育的水稻单片段代换系W11-09-02-07-02-03-01(W11-09-02-,Rf3Rf3/Rf4Rf4)对于野败型(WA)和矮败型(DA)细胞质雄性不育(CMS)系均具有较强的恢复性。单片段代换系W11-09-02-分别与WA-CMS和DA-CMS不育系组配,根据其BC3F2植株的花粉和小穗的育性,研究这2种不育细胞质的遗传关系、恢复基因Rf3和Rf4的遗传效应及单片段代换系W11-09-02-对这2种不育细胞质的遗传模式。【方法】选用WA-CMS、DA-CMS 2种细胞质的不育系博白A(BoA)和协青早A(XqA)为母本,以单片段代换系W11-09-02-为父本,采用单片段代换系W11-09-02-连续回交和分子标记辅助选择的方法,构建了2个具有单片段代换系W11-09-02-遗传背景的BC3F2群体,在这2个BC3F2群体中,筛选鉴定出携带基因型Rf3Rf3/Rf4Rf4、Rf3Rf3/rf4rf4、rf3rf3/rf4rf4和rf3rf3/rf4rf4的单株,并对其花粉和小穗的育性进行考察, 最后利用201个多态性SSR标记对这些单株进行遗传背景分析。【结果】在单片段代换系 W11-09-02-遗传背景下,WA-CMS的遗传效应大于DA-CMS,单片段代换系W11-09-02-中的恢复基因Rf4的遗传效应大于Rf3,而且这2个基因表现出累积效应。单片段代换系W11-09-02-中的恢复基因Rf3和Rf4,对WA-CMS和DA-CMS均表现出质量性状的遗传。【结论】DA-CMS的可恢复性优于WA-CMS,W11-09-02-中的恢复基因Rf4的遗传效应大于Rf3,这2个恢复基因对WA-CMS和DA-CMS表现出质量性状的遗传。

Analysis on restorer genes to male sterility of two different cytoplasmic in rice
Abstract:
【Objective】Rice chromosome single segment substitution line (SSSL) W11-09-02-07-02-03-01 (W11-09-02-,Rf3Rf3/Rf4Rf4),a strong restorer line for WA-CMS and DA-CMS,was recently identified at the State Key Laboratory for Conservation and Utilization of Subtropical Agro-bioresources,South China Agricultural University.In this study,based on the fertility in pollen and spikelet of the BC3F2 plants,the genetic relationship between WA-CMS and DA-CMS,the genetic effect and the genetic mode of the Rf3 and Rf4 genes for the two CMS systems were determined. 【Method】Two BC3F2 populations,possessing the genetic background of the SSSL W11-09-02-,were generated from the crosses between the SSSL W11-09-02-(recurrent parent) and the two CMS lines by backcrossing and marker-assisted selection.In the two BC3F2 populations,the plants carrying the Rf3Rf3/Rf4Rf4,Rf3Rf3/rf4rf4,rf3rf3/Rf4Rf4 and rf3rf3/rf4rf4 genotypes were selected,their phenotyping for fertility in pollen and spikelet were evaluated,and their genetic background was analyzed using 201 SSR markers.【Result】The genetic effects of WA-CMS were stronger than that of DA-CMS in the genetic background of SSSL W11-09-02-,and the restoration effects of Rf4 appeared to be slightly larger than that of Rf3,and their effects were additive for the two CMS systems.Two pairs of dominant genes governed the fertility restoration in pollen and spikelet in the SSSL W11-09-02-,indicating that the genetic mode of the Rf genes was a qualitative character for both WA-CMS and DA-CMS systems.【Conclusion】DA-CMS was restored more easily than WA-CMS.In the SSSL W11-09-02-,the effect of Rf4 appeared to be slightly larger than that of Rf3,and their effects were additive for the two CMS systems.The genetic mode of the two Rf genes was a qualitative character for the two CMS systems.
