真梅系梅花‘三轮玉蝶’叶片再生体系的初步建立

摘要:
【目的】初步建立真梅系梅花叶片的再生体系,为真梅系梅花良种培育及叶盘法基因转化等研究奠定基础。【方法】以真梅系梅花‘三轮玉蝶’叶片为试验材料,通过正交试验对外植体处理方法以及愈伤组织诱导、分化和生根培养基进行筛选,探讨真梅系梅花叶片再生的影响因素。【结果】 选用梅花‘三轮玉蝶’1年生枝条基部的叶片,背面向上接种并黑暗培养,有益于愈伤组织形成。在培养基1/2 MS+TDZ 2.0 mg/L+6-BA 0.5 mg/L+IBA 0.5 mg/L+NAA 0.1 mg/L中,叶片愈伤组织诱导率可以达到85.04%;愈伤组织转接在培养基1/2 MS+TDZ 1.0 mg/L+IBA 0.5 mg/L+NAA 0.2 mg/L中,分化率可达46.44%;分化出的不定芽在添加6-BA 0.5 mg/L+IBA 0.5 mg/L+NAA 0.3 mg/L的1/2 MS培养基上诱导生根效果较好,生根率达88.29%。【结论】明确了处理方法和部分外源激素对梅花‘三轮玉蝶’叶片愈伤组织形成的影响,初步建立了梅花‘三轮玉蝶’叶片再生体系。

Preliminary establishment of the regeneration systems for Prunus mume ‘Sanlun Yudie’ leaf-explants
Abstract:
【Objective】The regeneration system of Prunus mume leaf-explants was established to improve the research of genetic transformation and leaf disc.【Method】The leaves of ‘Sanlun Yudie’ were used as explants to explore the explants processing method,the optimal callus induction medium,the optimal callus differentiation medium and the optimal root differentiation medium in the orthogonal experiment.【Result】The leaves,from the annual twig base,back up in the medium and culturing in the dark,were conducive to the formation of callus.In the callus induction medium of 1/2 MS+TDZ 2.0 mg/L+6-BA 0.5 mg/L+IBA 0.5 mg/L+NAA 0.1 mg/L,the inducing rate was 85.04%.By culturing in the callus differentiation medium,1/2 MS+TDZ 1.0 mg/L+IBA 0.5 mg/L+NAA 0.2 mg/L,the differentiation rate was 46.44%.The root differentiation medium,1/2 MS+6-BA 0.5 mg/L+IBA 0.5 mg/L+NAA 0.3 mg/L,was taken as the proper result in the experimental analysis and the rooting rate was 88.29%.【Conclusion】Taking the leaves from P.mume‘Sanlun Yudie’ as explants,the effect of processing methods and exogenous hormones were defined basically.The regeneration system of P.mume ‘Sanlun Yudie’ leaf-explants was initially established.
