白鹃梅的离体培养及植株再生试验

摘要:
【目的】探讨不同植物生长调节剂对白鹃梅芽苗再生和生根的影响,为白鹃梅的开发利用、工厂化育苗及遗传育种研究奠定基础。【方法】以白鹃梅的嫩茎段为外植体,接种前用1 g/L的HgCl2溶液对材料进行不同时间(6,8,10,12 min)的消毒处理,筛选适宜的消毒时间;采用均匀设计法,将无菌外植体接种在附加有不同质量浓度植物生长调节剂(0,0.5,1.0,1.5和2.0 mg/L 6-BA,0,0.1,0.3,0.5和0.7 mg/L NAA)的MS培养基中,筛选诱导直接再生芽苗的适宜培养基;将直接再生芽苗接种在附加有不同种类和质量浓度植物生长调节剂(0.1,0.3,0.5和 0.7 mg/L 6-BA,0.1,0.3,0.5和0.7 mg/L IBA)的1/2MS培养基中,筛选诱导生根的适宜培养基。【结果】1 g/L HgCl2溶液对白鹃梅嫩茎段的最适宜消毒时间为8 min,在该条件下外植体的成活率为95.2%;白鹃梅直接再生芽苗的最适宜诱导培养基为MS+6-BA 1.0 mg/L,在该培养基上直接再生芽苗的诱导率可达85.7%;生根培养基以 1/2MS+ 0.3 mg/L NAA的生根效果最好,生根率达到70.45%。【结论】获得了白鹃梅嫩茎段适宜的消毒时间及适宜的再生芽苗和生根培养基配方。

In vitro culture and plantlet regeneration of Exochorda racemosa(Lindl.)Rehd
Abstract:
【Objective】The research of the effects of different kinds of plant growth regulator combination on bud differentiation and the influence on rooting effect were studied to set up a stable regeneration systerm and to lay a foundation and method for exploitation and application,Industrial Seedling,and the study of inheritance and breeding of Exochorda racemosa(Lindl.)Rehd.【Method】The tender stems of Exochorda racemosa(Lindl.)Rehd were used explants in the experiment.Explants were disinfected at various time(6,8,10,12 min) of using 1 g/L Hg solution before inoculated,and slected the best time for disinfection of explants;The explants disinfected were cultured in MS medium supplemented with different concentration of hormones(0~2.0 mg/L 6-BA,0~0.7 mg/L NAA) was filtrated and optimized by using uniform design;The shoots regeneration cultured in 1/2MS medium supplemented with different hormones and concentrations(0.1~0.7mg/L 6-BA,0.1~0.7 mg/L IBA)was filtrated and optimized.【Conclusion】The best time of disinfection in 1g/L Hg solution was 8 min,and the survival rate of explants was 95.2%;the best medium for shoots regeneration was MS+1.0 mg/L 6-BA,the rate of induction was 85.7%;the best medium for rooting was 1/2MS+0.3 mg/L NAA,the rate of rooting was 70.45%.【Result】It was obtained the tender stems of Exochorda racemosa needed a suitable time for sterilization,induction of the shoots regeneration by the tender stems and rooting of the shoots regeneration needed suitable concentration of formula of medium.
