不同来源乳酸杆菌生物学特性的比较

摘要:
【目的】比较不同来源乳酸杆菌的生物学特性,为益生菌及重组乳酸菌的宿主选择提供理论依据。【方法】采用CaCO3质量浓度为20 g/L 的CaCO3MRS培养基,从山羊瘤胃液和粪便、土鸡嗉囊和小肠壁附着物、青贮玉米秸秆、泡菜等不同样品中厌氧培养分离乳酸杆菌,对其耐热性、耐酸性、胆盐耐受性、抑菌性及耐药性、黏附性等生物学特性进行比较研究。【结果】共分离出了20株乳酸杆菌,其中7株含有内源性质粒,分别是SN1、JXH1、JXQ2、YF2、PC1、QZ5、QZ7,其耐热范围为37~60 ℃;其中PC1、JXH1具有较强的耐酸性;JXH1、JXQ2的胆盐耐受性为2~8 g/L,其上清液对大肠杆菌、金黄色葡萄球菌、蜡样芽孢杆菌均有抑制作用,对临床常用的氨基糖类、青霉素类、头孢菌素类和呋喃类抗生素敏感,对磺胺类、喹诺酮类、多肽类均有不同程度的耐药性;PC1、QZ5、SN1、JXQ2、YF2、JXH1对Caco-2细胞的黏附个数均为8.0~9.5个。【结论】在7株含有内源性质粒的乳酸杆菌中,SN1、JXQ2、YF2、JXH1对Caco-2细胞有较好的黏附性,同时表现出了较好的耐热、耐酸性和胆盐耐受性、抑菌性及对大多数抗生素的敏感性。

Contrastive study on the biologigal characterics of Lactobacillus from different sources
Abstract:
【Objective】This article was conducted to screen Lactobacillus from different samples and to study their biological characteristics,which provides a theoretical basis to select probiotics and ideal recombinant host bacteria.【Method】We collected samples by sterile operation and inoculated 20 g/L CaCO3 MRS,then isolated 5 genera of 20 Lactobacillus by anaerobic culture and studied the heat resistance,acid resistance,bile salt tolerance and adhesion of 7 strains with plasmid.【Result】The thermoresistance range of 7 Lactobacillus was 37-60 ℃;PC1 and JXH1 had powerful acid resistance;The bile salt tolerance range of JXH1 and JXQ2 was 2-8 g/L,and their supernatants had inhibitory action to 3 different pathogenic bacteria.The Lactobacillus,which contained Endogenous plasmid,were sensitive to amino sugar used in the clinic,penicillins,cephalosporins and furans antibiotics and had different levels of drug resistance to sulfonamides,quinolones and peptide;The adhesion number of PC1,QZ5,SN1,JXQ2,YF2,JXH1 was 8.0-9.5/Caco-2 cell.【Conclusion】 By studying the physiological characteristics of 7 Lactobacillus containing endogenous plasmid,we found that SN1,JXQ2,YF2,JXH1 of Caco-2 cell have better adhesion,and they also show better thermoresistance,acid and bile salt tolerance.Meanwhile they have activity of bacteriostasis and sensitivity to most antibiotics,which provides the theoretical basis and the original straint to the host for the recombinant lactic acid bacteria.
