黄土高原不同县域山杏种质遗传多样性研究

摘要:
【目的】研究黄土高原不同县域山杏种子可溶性蛋白质含量和电泳谱带的差异,为山杏种内分类及良种选育提供理论依据。【方法】采用形态学观测法,对黄土高原(陕西麟游和甘肃灵台、华池、庆城)山杏种质资源变异情况进行调查,从中筛选出形态类型典型的山杏,采用考马斯亮蓝G-250染色法和SDS-PAGE电泳法对种子可溶性蛋白质含量和谱带进行检测分析。【结果】共筛选出了37种山杏变异类型,其种子可溶性蛋白质含量差异较大。SDS-PAGE电泳分析表明,山杏种子可溶性蛋白质含量丰富、多态、稳定、谱带清晰,主要由41.1,39.0,34.6,22.7,22.0和21.7 ku 6种亚基组成,其余亚基数量丰富但含量较少。聚类分析表明,37种山杏类型可分为5个大类。【结论】种子可溶性蛋白质分类结果与形态特征和生物学特性存在一定的相似性,利用分类结果可对山杏品种进行改良。

Genetic diversity analysis on germplasm of Armeniaca sibirica in different counties in Loess Plateau
Abstract:
【Objective】The study was conducted to study the difference of content and electrophoresis bands on the soluble seed protein among different counties of Armeniaca sibirica (L.) Lam in broken Loess Plateau to provide theoretical basis for classification and breeding of A.sibirica.【Method】Morphological observation method was used to investigate the variations of A.sibirica.germplasm resources in broken Loess Plateau,and to sift typical morphotypes.The methods of Bradford and SDS-PAGE were used to analyze the soluble seed protein of A.sibirica.【Result】37 mutation types were sifted,the protein contents of which were quite different;The result of SDS-PAGE showed that soluble seed protein had such advantages as richness,stability,and polymorphism.The protein subunits band was clear,and the main components included six kinds of 41.1,39.0,34.6,22.7,22.0,21.7 ku,and the other subunits band was numerous but with few contents;The 37 mutation types can be classified into 5 groups.【Conclusion】The protein contents and electrophoresis bands are quite different,and the result has some similarity with the classification results on the morphological and biological characters.The classification results can be used for variety improvement of A.sibirica.
