【目的】 确定枯草芽孢杆菌溶血相关基因yqxC的功能。【方法】 采用PCR方法,扩增枯草芽孢杆菌溶血相关基因yqxC,构建GST融合表达载体,并在大肠杆菌中进行表达。使用含体积分数5%绵羊血的琼脂平板,检测可溶性融合蛋白GST-YqxC的溶血活性。此外,采用RT-PCR检测yqxC基因在枯草芽孢杆菌中的转录情况。【结果】 获得的可溶性融合蛋白GST-YqxC,在绵羊血琼脂平板上显示出较强的溶血活力;通过RT-PCR,检测到yqxC基因可以在枯草芽孢杆菌中转录。【结论】 枯草芽孢杆菌yqxC基因编码蛋白YqxC有较强的溶血活性,且能够发生转录,yqxC很可能是引起枯草芽孢杆菌溶血的基因之一。 |

Expression and functional characterization of a hemolysis associated gene yqxC from Bacillus subtilis |

【Objective】The study was done to identify the hemolysis associated gene yqxC fromBacillus subtilis.【Method】 A hemolysis-associated gene yqxC was amplified from the genomic DNA of B.subtilis by PCR.GST fusion expression vectors were constructed and fusion proteins were expressed in Escherichia coli.5% sheep blood agar plates were used to measure the hemolytic activity of the expressed soluble fusion protein.In addition,the transcription of yqxC was studied by RT-PCR. 【Result】 The soluble fusion protein GST-YqxC showed strong hemolyitc activity on sheep blood agar plates and yqxCwas indeed transcribed in B.subtilis.【Conclusion】yqxC encoded by yqxC in B.subtilis had the strong hemolyitc activity and yqxC was transcribed in B.subtilis.Therefore we infered that yqxCis possibly one of the hemolysin genes responsible for hemolysis in B.subtilis. |
