草莓试管内诱导匍匐茎和高温处理结合茎尖培养脱毒技术研究

摘要:
【目的】获得可用于生产的无病毒草莓种苗。【方法】在试管内对深山草莓花瓣愈伤组织分化产生的优良变异新品种的试管苗进行匍匐茎诱导,并采取试管内高温处理结合茎尖培养对该品种进行脱毒研究,应用均匀设计法对各步骤主要影响因子进行优化筛选。【结果】该草莓新品种试管苗匍匐茎诱导的适宜培养基为:LS+6-BA 1.00 mg/L+GA 3 1.90 mg/L +KT 1.00 mg/L,其诱导率为99.5%以上;将含有匍匐茎的试管苗在试管内于56 ℃条件下培养85 d后,切取1 mm的茎尖在培养基MS+6-BA 0.50 mg/L +NAA 0.20 mg/L中进行茎尖愈伤组织诱导,然后将茎尖愈伤组织转接到培养基MS+6-BA 1.00 mg/L +NAA 0.05 mg/L中进行愈伤组织芽苗再分化培养,对再生芽苗进行病毒检测,脱毒率为100%。【结论】利用试管内高温处理结合茎尖脱毒的方法可以完全脱除草莓病毒,从而建立了草莓脱毒体系。

Technology for stolon seedling in vitro of strawberry and virus eradication by combining the high temperature treatment with shoot tip culture
Abstract:
【Objective】The study was done to obtain seedlings without viruses which were used for strawberry producing.【Method】We induced stolon of variety with fine properties from petals of Fragaria orientalis Lozinsk.var.concolor (Kitag.) Liou et C.Y.Li in vitro,and studied virus eradication with combined shoot tip culture and high temperature treatment in vitro,the steps and the main factors of influence were screened through uniform design experiments.【Result】The medium of LS+6-BA 1.00 mg/L+GA 3 1.90 mg/L+KT 1.00 mg/L was suitable for stolon induction in vitro,with which the frequency of induction was more than 99.5%;after seedlingin vitro young stolon was cultured for 85 days at 56 ℃ in vitro,the medium of MS+6-BA 0.50 mg/L+NAA 0.20 mg/L was used for induced callus from shoot tip with 1 mm,after that callus of shoot tip was cultured on the medium of MS+6-BA 1.00 mg/L+NAA 0.05 mg/L for shoots differentiation,and virus detection with which the rate of virus eradication was 100%.【Conclusion】It is successful to get rid of viruses completely by combining the high temperature treatment with shoot tip culture in vitro,and the virus free system of strawberry has been established.
