真空微波预辐射辅助提取竹叶黄酮工艺的优化

摘要:
【目的】筛选真空微波预辐射提取竹叶黄酮的最佳提取工艺参数,为竹叶黄酮的提取和开发利用提供技术支持。【方法】将竹叶浸润,经微波预辐射辅助处理后进行水浴提取,利用响应面法得到二次多项式回归方程,研究浸润时间、微波功率和微波辐射时间及其交互作用对竹叶黄酮提取得率的影响,并在电镜下观察竹叶细胞的变化。【结果】真空微波预辐射辅助提取竹叶黄酮工艺的优化参数为:浸润时间60 min,微波功率17.23 W/g,微波辐射时间180 s,在此条件下,竹叶黄酮的提取得率为1.415%。电镜扫描结果表明,真空微波预处理竹叶的细胞结构受到较大程度的破坏,细胞破壁效果明显高于常规浸润预处理。【结论】真空微波预辐射有助于植物细胞壁的破壁,可减少后续浸提的时间和溶剂的消耗。

Optimization of extraction technology of vacuum microwave preradiation for flavonoids from bamboo leaves
Abstract:
【Objective】The extration technological parameters of flavonoids from bamboo leaves pretreated in advance by vacuum microwave were studied,which would provide theoretical basis and explore approach for extraction and development of flavonoid from bamboo leaves.【Method】Bamboo leaves were firstly infiltrated,then the microwave preradiation was conducted.The water bath extraction was doned for bamboo leaves.Investigated in the study were the effects of the wetted time,microwave power and microwave irradiation time on extraction rate of flavonoid from bamboo leaves.Single factor analysis and mutual effects of factors were analyzed by response surface methodology.【Result】 The optimal parameters were obtained as following:the soak time of 60 min,microwave power of 17.23 W/g,microwave irradiation time of 180 s,and the extraction rate up to 1.415%.【Conclusion】Vacuum microwave preradiation contributes to break for cell wall of plant,time of subsequent extraction and solvent consumption can be reduced correspondingly.
