人参种源遗传多样性和遗传关系的AFLP分析

摘要:
【目的】阐明不同种源人参的遗传关系和遗传多样性。【方法】用AFLP分子标记对来自我国吉林、辽宁2省及韩国、朝鲜的24份人参种源的多态性、亲缘关系和居群遗传多样性进行分析。【结果】从36对AFLP引物组合中,筛选出8对扩增条带清晰、多态性高的引物组合,用这8对引物共扩增出188个位点,其中138个位点为多态位点,多态位点百分率为73.4%;通过聚类分析,可将24个人参种源聚为4类;在集安、抚松和韩国的3个人参居群中,集安居群的Nei’s基因多样度、Shannon’s信息指数均最高,分别为0.168 9和0.273 8。【结论】AFLP分子标记研究结果能较好地揭示人参种源间的遗传多样性和遗传关系。

Genetic diversity and genetic relationship analysis of ginseng by AFLP Marker
Abstract:
【Objective】The research was to study the genetic diversity and genetic relationship of different resources of ginseng.【Method】Genetic relationship and genetic diversity in 24 accessions from Jilin and Liaoning province of China,Republic of Korea and Democratic People’s Republic of Korea were measured by using polymerase chain reaction (PCR) with AFLP Marker.【Result】Eight AFLP primers were selected from 36 AFLP primers and used for AFLP amplification.A total of 188 loci were generated,of which 138 loci were polymorphic loci (the percentage of polymorphic loci,P=73.4%).The similarity coefficients were calculated with NTsys-2.10 software and the dendrogram was constructed with UPGMA.A high genetic diversity was observed in Ji’an population among Ji’an population,Fusong population and Korean population with Nei’s diversity of 0.168 9 and Shannon’s inform index of 0.273 8.【Conclusion】Informative genetic diversity and genetic relationships of ginseng accessions were showed by AFLP Markers in the present study,providing a scientific basis for genetic breeding,differentiation and new cultivar selection.
