山羊KIF-Ⅰ基因外显子6与绒性状的相关性

摘要:
【目的】研究新疆3个地方山羊品种KIF-Ⅰ基因外显子6的多态性及其与产绒性状的相关性,为新疆山羊品种的选育提供依据。【方法】 利用PCR SSCP和DNA序列分析法,对213只新疆山羊、294只南疆绒山羊、253只博格达绒山羊的KIF-Ⅰ基因外显子6的多态性进行检测,并分析其与绒细度、绒厚度、产绒量和产绒后体质量的相关性。【结果】 ①KIF-Ⅰ基因外显子6区域存在1个突变点,即c.5106位发生了碱基T转变为C(T>C)的突变(M23912:c.5106 T>C)。在3个山羊品种中,检测到了TT、CC、TC 3种基因型,T和C 2个等位基因,其中T为优势等位基因,其在新疆山羊、南疆绒山羊、博格达绒山羊中的频率分别为0.502,0.509和0.583,且3个山羊品种均处于Hardy Weinberg 平衡状态(P>0.05)。②方差分析表明,该多态位点仅与绒细度存在着显著相关性(P<0.05),且CC基因型个体绒细度显著小于TC和TT基因型个体;绒厚度、产绒量和产绒后体质量在3种不同基因型间差异均不显著(P>0.05)。【结论】 CC基因型可作为山羊绒细度性状标记辅助选择的有效DNA标记。

Association of KIF-Ⅰgene exon 6 with cashmere traits of goat breeds
Abstract:
【Objective】 This paper was to study the association of polymorphism of cashmere goat KIF-Ⅰgene exon 6 with cashmere production traits so that it could provide a scientific basis for selective breeding.【Method】 PCR SSCP and DNA sequence method were used to detect polymorphism of KIF-Ⅰ gene exon 6 in 213 Xinjiang goats,294 Nanjiang cashmere goats and 253 Bogeda cashmere goats.Correlations between different genotypes and cashmere fineness,down cashmere thickness,cashmere yield,body weight after combing were analyzed.【Result】 ①The result showed the mutation M23912:c.5106 T>C in exon 6 of KIF-Ⅰ gene.Moreover,we detected three genotypes(TT,CC and TC),two alleles(T and C),the allele T was predominant in three populations.The frequencies of allele T in Xinjiang goat,Nanjiang cashmere goat and Bogeda cashmere goat were 0.502,0.509 and 0.583,respectively,which was at Hardy Weinberg equilibrium (P>0.05).②Moreover,there was a significant correlation between polymorphism and cashmere fineness at this locus.The individual mean fiber diameter for genotype CC was less than that of genotype TT and TC (P<0.05).However,in down cashmere thickness,cashmere yield and body weight after combing traits of CC,TC and TT genotypes of the correlation analysis,the results showed no significant difference (P>0.05).【Conclusion】 Hence,genotype CC of KIF-Ⅰ gene can be used as a molecular marker for breeding cashmere fineness in goat marker assisted selection.
