干旱胁迫下小麦叶片脱水素的表达与水分的关系

摘要:
【目的】 研究脱水素对小麦抗旱的作用,阐明脱水素表达与水分的关系。【方法】 以2个耐旱性不同的同核异质小麦品系(高抗和低抗)为材料,在盆栽自然干旱胁迫和复水条件下处理小麦幼苗,采用SDS-PAGE和Western blot方法,分析小麦在干旱和复水条件下叶片的相对含水量、细胞膜相对透性和脱水素表达量的变化。【结果】 随土壤干旱胁迫程度的加剧,小麦叶片相对含水量下降,细胞膜相对透性增大,有1条28 ku脱水素特异表达;耐旱性弱的小麦品系叶片中脱水素的表达早于耐旱性强的小麦品系,说明其对干旱胁迫更敏感;而耐旱性强的小麦品系叶片中脱水素蛋白含量高于耐旱性弱的小麦品系;复水后,小麦叶片相对含水量上升,细胞膜相对透性减小,脱水素蛋白在一小段时间内仍存在,说明其表达有一定的延续性。【结论】 小麦耐旱性与其受干旱影响的程度和脱水素的表达密切相关

The expression of dehydrin in wheat leaves under drought stress and its relationship with water
Abstract:
【Objective】 The function of dehydrin to drought tolerance, and the relationship between the expression of dehydrin and drouht were studied.【Method】 Two different drought resistant and isonuclear alloplasmic wheats(with strong tolerance and weak tolerance)were pot cultured under drought stress and rehydration.We analyzed the relationship between water content,membrane permeability and the expression of dehydrin in wheat leaves in the process of natural drought stress and rehydration.【Result】 Under drought stress,the water content of wheat leaves decreased,but membrane permeability increased.Western blot analysis showed that,there was a specific protein of 28 ku under drought stress,the expression of dehydrin in weak drought tolereant wheat leaf was earlier than that in strong drought tolerant wheat leaf,and the content of dehydrin in strong drought tolerant wheat was higher than that in weak drought tolerant wheat. After redehydration,the water content of wheat leaves increased,the membrane permeability decreased,and this dehydrin could exist in plant for some time.【Conclusion】 It shows that the harm to wheat under drought stress and the expression of this dehydrin are closely related to drought resistance of wheat.
