D-101大孔树脂分离纯化葛根异黄酮的工艺探讨

摘要:
【目的】 探索D-101大孔树脂分离纯化葛根异黄酮的最优工艺。【方法】 采用紫外分光光度法测定异黄酮质量浓度,以异黄酮损失率、洗脱率、收率、纯度等为指标,评价D-101大孔树脂分离纯化葛根异黄酮工艺中,上样液用量、上样液质量浓度、吸附流速、洗脱剂蒸馏水用量、洗脱剂乙醇体积分数及其用量对吸附和解吸效果的影响,从而确定最优工艺。【结果】 D-101大孔树脂对葛根异黄酮有较好的分离效果,其最优工艺条件为:葛根异黄酮饱和吸附量为3.3倍树脂体积,上样液质量浓度7.20 mg/mL,吸附流速为2 mL/min,洗脱剂蒸馏水和体积分数30%乙醇的用量均为4倍树脂体积。利用该工艺精制后葛根异黄酮纯度、收率分别达80.74%和63.62%。【结论】 采用D-101大孔树脂分离纯化葛根异黄酮简单可行,精制效果好,适于工业化生产。

Discussion on technique for separation and purification of Radix Puerarise isoflavonoids by D-101 macroporous resin

Abstract:
【Objective】 The study was to investigate the optimal technological parameters of the separation and purification process ofRadix Puerarise isoflavonoids by D-101 macroporous resin.【Method】 UV spectrophotometry was used to determine the content of isoflavonoids,and the effect of the work solution volume,the work solution concentration,the absorption velocity,the volume of distilled water and ethanol,the concertration of ethanol on the absorption and desorption of D-101 macroporous resin by the losing rate,eluting rate,recovery rate,purity of isoflavoniods were evaluated.【Result】 The D-101 macroporous resin had the best separating efficiency under the following technological conditions:the maximum absorption capacity of isoflavonoids was 3.3 times to the volume of macroporous resin;the work solution concentration was 7.20 mg/mL;the absorption velocity was 2 mL/min;the volume of distilled water was 4 times to the volume of macroporous resin;the concertration of ethanol was 30% (4 times to the volume of macroporous resin).The purity and recovery rate of the refined isoflavonoids was 80.74% and 63.62% respectively.【Conclusion】 This method is simple and feasible with good purification effect,which can meet industrial requirements.
