IFN-γ对妊娠早期大鼠下丘脑-垂体-卵巢轴中IGF-1表达的影响

摘要:
[目的]确定干扰素γ(IFN-γ)对妊娠早期大鼠下丘脑-垂体-卵巢轴中IGF-1表达的影响.[方法]将妊娠9 d的SD大鼠随机分为对照组、试验1组和试验2组,采用生殖道肌肉注射的方法,对照组大鼠生殖道肌肉注射生理盐水,试验组大鼠注射IFN-γ,其中1组剂量为2.5×104U/只,2组剂量为7.5×104U/只.各试验组均在注射48h后灌注取材、免疫组化SP法染色,光镜下观察.[结果]注射2.5×104U/只和7.5×104U/只的IFN-γ均能下调妊娠大鼠下丘脑室周核、视上核、视前大细胞核、视前内侧核、视前外侧核、视前交叉上核、弓状核,腺垂体,卵巢黄体及子宫蜕膜基质细胞和子宫腺中IGF-1的表达.[结论]IFN-γ对妊娠早期大鼠下丘脑-垂体-卵巢轴中IGF-1的表达具有下调作用,高剂量IFN-γ的下调作用更显著.

Effects of IFN-γ on the expression of IGF-1 in Hypothalamus-Pituitary ovary-Gonad axis of early pregnant rats
Abstract:
【Objective】 The study investigated the effect of interferon-γ(IFN-γ)on the expression and distribution of insulin-like growth factor-1(IGF-1)immuonorea-ctive production in Hypothalamus Pituitary ovary-Gonad axis of earlier pregnant rats in the post-implantation period.【Method】 In this study the ultrasensitive immunohistochemistry SP method was used 9d pregnant rats were divided into control group,experimental group one and experimental group two.The control group was injected physiological brine and the two experiment groups were injected 2.5×104U and 7.5×104U of IFN-γ in vaginal muscular respectively.Experiment materials were collectd after perfusion from each group which was injected 48h ago,then stained by ultrasensitive immunohistochemistry SP method.And the results were observed and analyzed.【Result】 The expressions of IGF-1 in pre-opticus medianus,nucleus supra-opticus,nucleus pre-opticus mango cellularis,nucleus pre-opticus medialis,mucleus pre-opticus lateralis,nucleus pre-opticus suprachiasmatic and arcuate nucleus of the hypothalamus, adenohypophysis,corpus luteum of ovary,stroma cells of uterus decidua and uterine gland were reduced after being injected 2.5×104U and 7.5×104U IFN-γ.【Conlusion】The results suggested that IFN-γ could reduce the expression of IGF-1 immunological reaction production in Hypothalamus-Pituitary gland-Gonad axis.High dose of IFN-γ showd a much more significant down regulation.
