毛毡杜鹃离体培养及种质试管保存体系的建立

摘要:
[目的]建立毛毡杜鹃离体培养及种质试管保存的适宜体系.[方法]以毛毡杜鹃新生嫩芽为外植体,应用均匀设计法筛选其最适合的基部直接再生芽苗培养基、生根培养基及种质试管保存培养基.[结果]毛毡杜鹃最适合的基部直接再生芽苗诱导培养基为:DR+ZT 3.00 mg/L+IAA 0.03 mg/L,分化率达100%;生根培养基为:MS(改良)+IAA 0.10 mg/L+IBA 0.10 mg/L+NAA 0.05 mg/L+KT 0.10 mg/L,生根率达97%以上;试管保存培养基为:1/10MS+B9 3.50 mg/L+根皮苷2.60 mg/L,保存时间可达38个月以上.以再生植株茎节为材料进行快繁的试验结果表明,在30 d培养周期内,每段增殖倍数平均达6倍以上.在常温条件下,采取"低营养矮化延缓生长"的方法在试管内保存毛毡杜鹃种质,成功地建立了毛毡杜鹃的离体培养和种质试管保存体系.[结论]所建立的离体培养体系及种质试管保存体系可以满足毛毡杜鹃离体繁殖和种质保存的需要.

In vitro culture and germplasm peservation in vitro system of Rhododendron confertissimum Nakai.
Abstract:
【Objective】 In vitro culture and germplasm peservation In vitro system of Rhododendron confertissimum Nakai was established. 【 Method】 The tender buds of Rhododendron confertissimum Nakai.were used as explants for the experiment. Uniform design for the most suitable media for shoots regeneration immediately at base of tender buds,rooting and germplasm preservation In vitro was screened. 【Result】 The results showed that DR+ZT 3.00 mg/L+IAA 0.03 mg/L was suitable for shoots regeneration,the rate of regeneration was 100%;MS(modified)+IAA 0.10 mg/L+IBA 0.10 mg/L+NAA 0.05 mg/L+KT 0.10 mg/L for rooting,the rate of rooting was more than 97%;1/10MS+B9 3.50 mg/L+Phloridzin 2.60 mg/L for germplasm preservation In vitro for 38 months.Stems,each with one node,were cut from regenerated shoots and cultured for propagation,and a 6-fold proliferation rate was achieved within 30 days.The method of “low nutrients with defering growth and dwarfing” was utilized for germplasm peservation in vitro at normal temperature.
