太白蓼提取物体外抑制新城疫病毒及5种细菌的活性部位筛选

摘要:
[目的]筛选太白蓼提取物抑制新城疫病毒(NDV)及5种细菌的活性部位.[方法]采用细胞病变法和MTT法测定太白蓼提取物对NDV在鸡胚成纤维细胞(CEF)上增殖的影响,并用琼脂扩散和试管稀释法进行了抑菌试验.[结果]太白蓼乙酸乙酯部位(PTKE)及其分离物EB,对NDV在鸡胚成纤维细胞(CEF)上的抑制率分别为32.49%和66.31%(P<0.01);分离物EB药毒混合感作组对NDV的抑制率显著高于先给药后接毒组和先接毒后给药组(P<0.01),质量浓度在78.13 μg/L以下时,先给药后接毒组对NDV的抑制率显著高于先接毒后给药组(P<0.01),当质量浓度为156.3 μg/L以上时,2组间差异不显著(P>0.05),药物质量浓度与病毒抑制率间存在明显的量效关系(P<0.05).PTKE对大肠埃希菌、金黄色葡萄球菌、沙门氏菌、多杀性巴氏杆菌和无乳链球菌的最低杀菌浓度(MBC)分别为100,25,50,25 和50 mg/mL;而PTKE 的分离物EA、EB和EC对大肠埃希菌和巴氏杆菌无抑菌作用,EA对金黄色葡萄球菌、沙门氏菌的MBC分别为2.5和5.0 mg/mL,EB对金黄色葡萄球菌、沙门氏菌和无乳链球菌的MBC分别为1.25,5.0和2.5 mg/mL,EC对金黄色葡萄球菌的MBC为2.5 mg/mL.[结论]太白蓼提取物具有体外抑制NDV和细菌的作用.

Selection on active sites of Polygonum taibaishanense Kung.extratives inhibiting NDV and 5 bacteria in vitro
Abstract:
【Objective】 The experiment selected the active site of Polygonum taibaishanense Kung.extratives on inhibiting NDV and 5 bacteria. 【Method】 Cellular pathological change and MTT methods were used to study the proliferation and inhibition of Polygonum taibaishanense Kung.extratives on chick embryo fibroblast(CEF),and antibacterial effect was observed by agar diffusion and test dilution methods. 【Result】 The inhibition rate of ethyl acetate site (PTKE) and isolation EB in Polygonum aibaishanense Kung.on NDV in CEF were 32.49% and 66.31% respectively.The inhibition rate of EB on NDV in drug virus mixed infection group were significantly higher that in NDV infected group which was firstly prevented by drugs and drug-treated groups which were firstly infected by virus (P<0.01).The inhibition rate of EB on NDV in group 2 was significantly higher than that in group 3 when the dose was lower than 78.13 μg/L (P<0.01),no difference was observed between the two groups when the dose was higher than 156.3μg/L (P>0.05).Test of different doses showed a kind of obvious dose-effect relation (P<0.05).The minimal bactericidal concentration (MBC) of PTKE on Escherichia coli,Staphylococcus aureus,Salmonella,Pasteurella multocida and Streptococcus agalactiae were 100,25,50,25 and 50 mg/mL respectively.The isolation of EA,EB and EC had no antibacterial effect on Escherichia coli and Pasteurella multocida.MBC of EA on Staphylococcus aureus and Salmonella were 2.5 and 5 mg/mL,and MBC of EB on Staphylococcus aureus,Salmonella and Streptococcus agalactiae were 1.25,5 and 2.5 mg/mL respectively,while MBC of EC on Staphylococcus aureus was 2.5 mg/mL.【Conclusion】 Polygonum taibaishanense Kung.Extratives have inhibition effect on NDV and several bacteria in vitro.
