秦川牛H-FABP基因第1外显子SNP及其与部分肉用性状相关性的研究

摘要:
[目的]分析秦川牛心脏脂肪酸结合蛋白基因(H-FABP)第1外显子的遗传变异与部分肉用性能指标的相关性.[方法]随机选择相同饲养条件下的146头18~20月龄秦川牛阉牛,采用PCR-SSCP结合测序技术,对秦川牛H-FABP基因第1外显子的遗传变异进行检测;运用SPSS统计程序中的GLM模型,分析其与部分肉用性能指标的关系.[结果]秦川牛H-FABP基因第1外显子130 bp处具有PCR-SSCP可检测到的SNP位点:Y=T→C.对该SNP位点具有不同基因型的个体进行分型并与88头秦川牛的肉用性状指标进行GLM分析,表明AB型个体的后腿围、背膘厚、大理石花纹等性状极显著高于AA型个体(P<0.01),AB型个体的胴体胸深和嫩度显著高于AA型个体(P<0.05),AB型个体的背膘厚极显著高于BB型个体(P<0.01);另外,BB型个体的后腿围、大理石花纹等性状极显著高于AA型个体(P<0.01).[结论]秦川牛H-FABP基因第1外显子上存在SNP位点且与肉用性状有较强的关联性,说明H-FABP基因可作为秦川牛部分肉用性状的候选基因.

Study on SNP of H-FABP gene extron 1 and its association with some meat traits in Qinchuan cattle
Abstract:
【Objective】 The association between hereditary variation of H-FABP gene extron 1 and some meat traits was analyzed.【Method】 146 18-20-month-old Qinchuan bullocks under identical feeding condition were selected randomly and part section hereditary variation ofH-FABP gene was analyzed by PCR-SSCP technique.The association between SNP locus and some meat traits was analyzed using the general linear model (GLM) in SPSS program.【Result】 The results showed that there was a mutation(Y=T→C)at 130th bp ofH-FABP gene extron 1 in Qinchuan cattle.Different genotypes of SNP locus were grouped and its relationship with meat traits of 88 Qinchuan cattles was analyzed using the general linear model.The crural girth,back-fat thickness,and marbling of AB genotype were significantly different from those of AA genotype (P<0.01).A significant diference was found between the genotypes of AB and AA for carcass depth and tenderness (P<0.05).There was a difference between the genotypes of AB and BB for back-fat thickness (P<0.01),as well as between the genotypes of AA and BB for crural girth and marbling (P<0.01).【Conclution】 SNP locus of H-FABP gene extron 1 in Qinchuan cattle is associated strongly with some meat traits.It indicates that H-FABP gene may be a candidate gene for some meat traits of Qinchuan cattle.
