英法两国不同生理小种苹果黑星病菌遗传多样性的SSR分析

摘要:
[目的]对来自英、法两国20株苹果黑星病菌的遗传多样性与其生理小种和地理来源之间的关系进行研究.[方法]设计了5对SSR引物,对分属于7个生理小种的苹果黑星病菌的基因组DNA进行了PCR扩增,检测其多态性位点,计算各位点的多态性信息含量(PIC)和菌株间的遗传相似系数(GS),并对各菌株间的亲缘关系进行了聚类分析.[结果]5对引物共检测到43个等位位点,每对引物平均获得等位位点8.6个;PIC变幅为0.76~0.90,平均为0.85,GS变幅为0.72~1.0,平均为0.85;供试苹果黑星病菌的遗传多样性与其生理小种和地理来源之间均无明显相关性.[结论]英、法两国的苹果黑星病菌之间存在人为原因造成的基因流动,抑制了自然发生的遗传变异;病菌生理小种的划分应以传统生物学方法为主.分子生物学分析方法为辅.

Genetic diversity of different races of Venturia inaequalis from UK and France with SSR markers
Abstract:
【Objective】 Genetic diversity of 20 isolates of Venturia inaequalis of 7 physiological races from UK and France were tested so as to find out the corelation between genetic diversity of Venturia inaequalis and their sources and races.【Method】 5 primer pairs of SSR markers were designed for PCR,the diversity loci,polymorphic information content(PIC),and genetic similarity(GS) were calculated and cluster analysis was tested.【Result】 43 alleles were detected with an average of 8.6 alleles per SSR primer pair,PIC was from 0.76 to 0.90 with an average of 0.85,GS was from 0.72 to 1.0 with an average of 0.85,genetic diversity of 20 isolates of Venturia inaequalis had minor relevance with their races and sources.【Conclusion】 The gene flows caused by human activity between Venturia inaequalis of UK and France had inhibited their naturally occurring genetic diversity;the traditional biological methods should be used in categorizing Venturia inaequalis races as well as molecular biology methods.
