[目的]了解两藏披碱草属野生牧草的遗传多样性及其数量、分布规律.[方法]以披碱草基因组DNA为模板,利用微卫星(Microsatellite)分子标记对搜集到的9份西藏披碱草属野生牧草及8份对照材料进行遗传多样性研究,统计了不同引物对供试材料的扩增结果,分析了西藏的9个披碱草居群间的遗传距离、遗传相似系数,并对所有17个居群进行了UPGMA聚类.[结果]试验共筛选32对引物,其中14对引物有扩增条带,具有多态性且扩增效果较好的引物有8对.用筛选出的8对引物对供试的17份材料进行微卫星分析,共检测出69个等位基冈,平均每对引物所产生的等位基因数为8.6个;西藏9个居群间的遗传相似系数为0.273~0.900.UPGMA聚类分析表明,在相似系数阈值为0.58时,9份材料可大致分为3类.[结论]西藏披碱草属野生牧草种质资源具有较高的遗传多样性. |

Abstract: |
【Objective】 The study was done in order to know the genetic diversity,quantity and distribution of the wild Elymus L.of Tibet germplasm resources.【Method】Elymus L.genome DNA was used as template,genetic diversity of 9 Tibet populations with 8 control groups were studied by microsatellite method.The amplified results by different primers were recorded,the genetic distance analyzed,Nei's genetic identity of 9 Elymus L.populations of Tibet analyzed and UPGMA Dendrogram cluster analysis of 17 Elymus L.populations made.【Result】 Thirty-Two primer pairs were used for polymorphic selection,with14 primer pairs obtained amplified products,eight primer pairs produced specifically polymorphic products.A total of 69 alleles were observed,the average number of alleles per locus was 8.6.Genetic identity ranged from 0.27 to 0.90 among the 9 populations.Cluster analysis with UPGMA showed that when genetic identity was 0.59,the 9 accessions could be divided into 3 groups.【Conclusion】 The wild Elymus L.of Tibet germplasm resources had a high genetic diversity. |
